The Chinese medicine YZ is used as a hemostatic medicine in our country, it’s dried bark can stop bleeding, so it has well foreground of exploitation.The subject is to screen out the active fractions, the drug toxicity, and the hemostatic mechanism from YZ by pharmacodynamics experiments, acute toxicity test,and coagulation tests. In addition, in order to determine the effective initial parts of the main chemical ingredient, we also carried out a qualitative chemical composition tests for the effective parts of YZ.Using rabbit femoral artery bleeding test and hemostasis test in rat liver to stop bleeding with YZ powder, screening the active fraction of hemostasis from YZ system extracts by clotting time and bleeding time test in mice. Results:The powder of YZ as the same as Yunnanbaiyao can significantly declined the bleeding time of rat liver and rabbit femoral artery; Compared to the control group, the high doses 20.00g/kg(crude drug) and the medium doses 10.00g/kg(crude drug) of petroleum ether extracts can obviously shortern mice CT & BT (p<0.01). Compared to Yunnanbaiyao group, the high doses can decreased CT of mice, there are significantly different (p<0.05), the curative effect is better than Yunnanbaiyao group; The high doses 20.00g/kg (crude drug) of chloroform extracts can significantly shorten CT& BT (p<0.05); By reversed-phase extraction method for petroleum ether extract of YZ, we research the effects of water-soluble part of petroleum ether extract on CT & BT in mice, the result showed that water-soluble part can shortern CT & BT significantly(p<0.01). Therefore, the Chinese medicine YZ can stop bleeding, the water soluble part of petroleum ether extract is the active fraction for hemostasis.We did not get the LD50 through the oral acute toxicity of the active fraction on mice. By maximum tolerated dose method, the dose 320.00g/kg (crude drug) was given to mice, there was not yet observed the apparent toxic effects. According to toxicity grading standards, it is non-toxic.The active parts were analyzed by chemical identification, the results showed that it mainly contents steroids, flavonoids and terpenoids.In order to study the hemostatic mechanism of active parts, the experiments of testing PT, APTT, TT and FIB were measured. Results showed that the high dose 20.00g/kg(crude drug) of WPEY can decreased APTT and TT apparently (p< 0.05), the high dose 20.00g/ kg (crude drug) can increased FIB obviously (p< 0.01), the medium dose 10.00g/ kg(crude drug) also can significantly increased FIB (p< 0.05). YZ can play a role in haemostasis and coagulation by affecting the intrinsic pathway of coagulation and fibrinolytic system.
