Barley is one of the four major food crops in the world, it is a typical "five yuan" crops (grain crops, forage crops, economic crops, health and energy crop), also has a variety of function compound, such as (β-glucan, γ-aminotutyric acid, flavonoids, Resistant starch and so on, catechin, myricetin, quercetin, kaempferol mainly flavonoids with elimination of oxygen free radical, antioxidant, antibacterial, anti-inflammatory, antiallergic, anti mutation, anti-tumor, protect the cardiovascular disease and so on. The important agronomic and quality of barley are generally controlled by many gene and interaction, also specific expression in different environment, find control complex quantitative trait gene by QTL anysis and guide the breeding through near molecular markers. The results show that:1 the optimum of HPLC method and extraction process of the major flavonoids in barley through orthogonal regression, HPLC method, Agilent Zorbax Eclipse XDB-C18(5μm. 250mm-4.6mm id) column; mobile phase A-1% glacial acetic aqueous, B-acetonitrile, gradient elution program:0~10min,15%~30% B; 10-16min,30%~40% B; 16~ 20min,40%~-15% B; flow rate 0.8ml/min, column temperature 30℃; detection at 280nm. injection volume of lOμL. The optimum ultrasound condition was as follows:methanol concentration.50%. soild-liquid ratio.1:20. and ultrasound time,60min.2 A total of 51QTLs were detected on 7 chromosomes include 31 additive QTLs and 20 epistatic QTLs.10 additive QTLs and 5 epistatic QTLs relate to four kinds of flavonoids; 21 additive QTLs and 15 epistatic QTLs relate to four agronomic traits. QTL distribution on the 7 chromosomes at most.21, on the 1 chromosomes at least, only 1 while the 2H,3H. 4H,5H and 6H chromosomes were 14,8.9 and 2. In this study, Seven QTL clusters involving more than one trait were detected, which were localized on chromosomes 2H, 3H,4H and 7H, on the 7 chromosomes at most,3, on the 3 chromosomes at least, only 2, while the 2H and 4H chromosomes were 1, this experiment considering QTL and invironment interaction, we detected 1 additive QTL and 6 epistatic QTL were with significant environmental effects.
