| Cerasus cerasoides var. cerasoides is larger in the development and utilization of wild Cerasus resources in value, the only in late autumn to early spring flowering ornamental cherry blossoms species,but its seed germination rate is extremely low, under natural conditions at present has not yet been development and utilization in garden.With the explants of of Cerasus cerasoides var. cerasoides, we studied the selection of basic medium, the stem section as explant during different growth regulating substances in single bud induction, cluster bud proliferation, take root and leaf callus induction, proliferation process function, to establish a high pot cherry regeneration system, for its large-scale application in city landscaping to improve technical guarantee.The results of the experiments are as follow:1.The best time in Cerasus cerasoides var. cerasoides explant is in early February.2.The best sterilization methods of explants of Cerasus cerasoides var. cerasoides:20 seconds is good to treat the Stem section by C2H5OH(70%),300 seconds is good to treat the Stem section by HgCl2(0.1%).3.The beginning of adventitious bud induction generation culture of Cerasus cerasoides var. cerasoides:Based on the impact factors onthe explants, basal medium, plant growth regulator concentrations and ratios, the optimalinduction medium was confirmed as MS+6-BA1.0mg/L+IBA0.1mg/L, the budding rate of the stem section was82.2%.4.The Subculture of Multiple shoot clumps:Studied 6-BA, IB A, NAA, the influence of different concentration combinations of clump buds transgenerational proliferation, one of the most significant influence on transgenerational proliferation effect is 6-BA, the optimalinduction medium was confirmed as 6BA0.8 mg/L+IBA 0.06 mg/L +NAA0.08 mg/L, the proliferation rate was 7.32, and growth in good condition, leaf color green.5. Seedling culture:The plants grown well and health in the MS medium without any plant growth regulators6. Rooting induction culture:Studied the basic culture medium and the concentration of IBA ratio effect on rooting and the most suitable medium for the rooting of the renewable effective seedlings of Cerasus cerasoides var. cerasoides was 1/2MS+IBA 0.5 mg/L, with the rooting rate was 92.22%, and the average number of rooting was 6.47,the length of root was 4.8(cm).7. Hardening seeding and transplanting:the optimized transplant medium for Cerasus cerasoides var. cerasoides was river-sand,humus mixed and vermiculite by 1:1:2, which the rate of survival was highest, Up to 72.7%, and the growth is very well.8.Blades induced callus culture of Cerasus cerasoides var. cerasoides:MS+6BA 1.0mg/L +NAA0.5mg/L is the best medium to induce callus,explant materials of the highest rate,81.11%, but Cerasus cerasoides var. cerasoides callus callu s stopped growing in the end. |
