In this paper, through the study of the shape of Sichuan tobacco area, the content of aroma, enzyme activity, as well as key metabolism key enzyme gene expression analysis, the conclusions are as follows:1.Flue cured tobacco terpenoid metabolic differences in various regions of SichuanIn flue cured tobacco leaves volatile matter content in different regions were significantly different.. The content of Honghuadajinyuan in Miyi area was the highest. The Mianning area was the lowest, and the Miyi area was more than two times. Yunyan 87 in Luzhou area was the highest, the Mianning area was the lowest, a difference of nearly two times. Volatile matter content of neophytadiene is the largest amount of a single substance, and neophytadiene content directly affects the total volatile organic compounds.The growth and development of flue-cured tobacco in different regions in the process of Lipoxygenase (LOX) enzyme activity are different in different periods, in most areas the growth of flue-cured tobacco in the process of lipoxygenase activity reached the maximum at budding stage.The comprehensive analysis we can get in the late growth stage of flue-cured tobacco of Lipoxygenase (LOX) enzyme activity in flue-cured tobacco is strong, high and volatile matter content; lipoxygenase (LOX) enzyme activity in flue-cured tobacco is weak, low volatile matter content.The growth and development of flue-cured tobacco in the process of the relative expression levels of LOX mRNA were different with different. And in the tobacco growth process showed increase first and then decrease, generally in most areas of the budding stage LOX gene relative expression was the maximum. We can get the relative expression of LOX gene, the direct effects of Lipoxygenase (LOX) activity. The relative expression of LOX gene is high, Lipoxygenase (LOX) enzyme activity is strong; the expression of LOX is low, the lipoxygenase (LOX) enzyme activity.2.Flue cured phenolic metabolism differences in various regions of Sichuan The total polyphenols have significant differences in different regions. Polyphenol content in flue-cured tobacco growth later period accumulated more.Mianning Yunyan 87 highest total polyphenol, polyphenol content was the lowest in Miyi Yunyan 87.And Hongda polyphenol content is high in Miyi, Huili area Honghuadajinyuan polyphenol content was the lowest.Phenylalanine ammonia lyase (PAL) and cinnamic acid -4- hydroxylase (CA4H) are involved in the synthesis of two enzymes of polyphenols. which belongs to the two key rate limiting enzyme in the same way.In all times and places of two kinds of enzyme activity have significant differences, but differences in the key period of polyphenol content in flue cured tobacco leaves is the enzyme activity level of maturity, as phenylalanine ammonia lyase (PAL) and cinnamic acid -4- hydroxylase (CA4H) enzyme activity is higher in mature period, in flue cured tobacco leaves the higher the content of polyphenols.Differences between flue-cured tobacco PAL gene relative expression in all times and places. And showed a decreased trend in tobacco growth process, but the PAL gene high expression under normal conditions the higher PAL activity, the contents of polyphenols in flue-cured tobacco was higher. The growth of flue-cured tobacco was different during different periods of relative expression of CA4H gene, CA4H gene relative expression quantity in each period has different values. CA4H gene is the key in the process of polyphenols metabolism genes.3.Tobacco alkaloids metabolic differences in various regions of SichuanThe area still exist significant difference in nicotine content.The highest content of nicotine of Hongda in Luzhou, the lowest in the nicotine content of Huili; Yunyan 87 in nicotine content of Guangyuan is the highest, in Miyi and Huili content is low. Most areas have at least one variety of nicotine content in the appropriate.But the content of nicotine in flue-cured tobacco Huili is low, while Guangyuan and Luzhou have a higher content of nicotine.There exist obvious differences in gene content of flue-cured tobacco in different regions of relative expression of PMT. The relative expression of PMT gene in different regions of Flue-cured Tobacco under normal circumstances the first increase in the growth of flue-cured tobacco in the process and then decreased, reached the maximum at squaring stage. Results PMT gene relative expression quantity of high nicotine content in the region is relatively high. The relative expression of PMT gene is relatively low, the nicotine content area is relatively low.
