Burn generally refers to tissue damage by heat(including hydrothermal, steam, high temperature gas, flame, energy, chemicals, radiation, burned metal liquid or solid, etc).For frequent burns, studying of traditional Chinese medicine was also deepening.The main subject was to observe the protective effect of Jinguolan ointment on the burn wounds in rats, and to further explore the mechanism to provide a solid theoretical foundation for further promoting the development of clinical applications and new formulations of Jinguolan ointment.The main experiments and results were as follows:1. Shallow second degree burns established on rat modelRat back hair was removaed after 24h of immersion in boiling water,100g weights respectively applied to the skin of rats for 5s,10s,15s and 20s.24h later scald burn wound full-thickness skin were excised for biopsy, then observe the skin epidermis and dermis pathological changes, finally scald 5 s could establish a stable shallow degree burns in rats.2. Effect of jinguolan ointment on burn wound healing in rat model of shallow degree burns.20 SD rats, male and female, were divided into 5 groups of four.30min later scald burns in each group were wiped drug in order to evenly cover the wound, bandaged by sterile gauze, administered once a day. Continuous administration until the wound was completely healed. Model group were given treatment ointment. Healing time and healing rate of each time were recorded at different points in time.Epithelium covering the wound was the basis of completely healed. Compared with the model group, the high-dose group (P<0.01) and middle dose group (P<0.05) could significantly shorten the healing time. After injury 7d, compared with the model group, high dose group and middle dose group were significantly higher healing rate (P<0.05), this trend had continued until injuried 14d.3. The possible action mehanism of jinguolan iontment on rat model of shallow degree burns84 SD rats, male and female, the normal group had 4, the remaining rats were randomly divided into 5 groups. After burning, the groups were given medication, after injury 1,3,7,14d, each group was randomly selected four rats, male and female, tail vein blood serum from experience, cut the wound tissue and stored at -80℃ refrigerator.By observing the rats after scald 1d-14d of histological changes and measuring physiological indicators,we discovered these as follows:The first three days after the injury, the IL-6 levels of serum rised significantly and reached a peak, the difference between the normal group and experimental groups were significantly (P<0.01), which shows successful model. From SOD and MDA contents of the wound tissue.,we can see that in the early burn wound, organism had a deepening process. Hydroxyproline content of the drug groups after the first three days of the injury reached the peak, earlier than the model group, and the peak concentration was significantly higher than the model group.In the first 7d, SOD activity of high, medium and low dose group were significantly higher than model group(P<0.01).At the same time,MDA content of each group were maintained downward trend, a large decrease in the high-dose group, significant differences with the negative group (P<0.05).EGF levels peak time of each drug group was earlier than the model group,and in the first 7d,Compared with the model group, positive control group (P<0.01), high-dose group (P<0.01) and dose group increased significantly (P <0.05).After injury 7d, IL-6 levels of each group decreased with time, compared with the model group, the high-dose group (P<0.01) and low-dose group (P<0.05) decline were more pronounced. At the same time,the serum content of TGF-β reached the peak, the high dose group increased significantly compared with the model group, the difference was significant (P<0.01). Jinguolan ointment could avoid further deeping the depth of burns in early time,lessen infiltration of inflammatory cells, accelerate wound capillary fiber nuclei, promote skin covering, and ultimately achieve the purpose of promoting wound healing.Conclusion:1.Jinguolan ointment could promote rat shallow second degree burn wound healing, improve wound histopathological changing.2.1t had a good protective effect on shallow second degree burn model in rats.3.The mechanism may involve promoting wound fibroblast growth, inhibition of lipid peroxidation, increasing antioxidant capacity wounds, promoting cell proliferation and the development of granulation tissue, controlling over part of the synthesis and releasing of inflammatory mediators.
