As one of the oldest fruit trees, grapes have important economic value. Great progress has been made in conventional cross breeding, but grape lags behind other economic crops in molecular genetic map construction and marker assisted breeding in China. In this study, 88 progenies derived from ’Wink’(Vitis vinifera L.) selfing were used as mapping populations (’WS’population), and the molecular genetic linkage map based on SSR markers was constructed for’WS’population. The Bulked Segregate Analysis (BSA) technique was applied to localize the male-sterile gene on the map and the bioinformatics was used to analyze the information of regions associated with this gene. Besides, the QTLs conferring leaf morphology and two phonological traits were located on the genetic map.The main results are as follows:1. A set of 470 SSR and 180 EST-SSR primers collected from NCBI and some literatures were screened in ’Wink’ and seven selfing individuals. OF them,141 SSR and 28 EST-SSR yielded polymorphic ampliations. These markers were used to analyze the genetype of mapping population, which consisted of 88 progenies from a selfing of V. vinifera’Wink’. By using Joinmap 4.0 software, the genetic map was constructed and contained 21 linkage groups filled with 134 SSR and 27 EST-SSR. The total length of the map was 1333.3cM, with an average distance of 8.28cM between markers.2. To map the male sterile gene in grape, a’WS’population including 88 individuals derived from the selfing of ’Wink’ was used as materials for gene mapping. In order to locate the gene and carry out bioinformatics analysis, the bulked segregate analysis (BSA) technology was conducted to screen the polymorphism between the fertile and sterile pools by SSR technologies. Two SSR markers of VVMD34 and VVIB23 were linked with VvMsl. They located on either sides with the genetic distances of 3.5cM and 1.9cM, respectively. The physical distance between VVMD34 and VVIB23 is 1134kb based on the whole genome sequence of grape, and there are 111 candidate genes in this region. These results will benefit fine-mapping of male-sterile gene and marker-assisted selection (MAS) in grape breeding program.3. Use of interval mapping allowed the identification of QTLs of grape by MapQTL5.0 sofeware. Twenty-seven QTLs for leaf morphology traits were detected and named from lmc-1 to lmc-27 in turn. These QTLs were located in LG11, LG17, LG16-1, LG14 and LG2, with the phenotypic variance from 12.5% to 40.5%. Four QTLs for leaf-expansion period were detected and named from lep-1 to lep-4 in sequence. These four QTLs were located in LG10 and LG7, with the phenotypic variance from 12.4% to 14.4%. Three QTLs for full-blossom period were detected and named fbp-1,fbp-2 and fbp-3. These three QTLs were located in LG14and LG2, with the phenotypic variance of 12.7%、13.3% and 13.8%, respectively。...
