| Abstract Paulownia witches’ broom is a contagious disease caused by phytoplasma, not only causing huge economic losses in paulownia production, but also seriously affectting the enthusiasm of the people to plant paulownia. Plant total DNA methylation level is too high or too low can lead to abnormal plant morphogenesis. Paulownia witches broom seedling DNA methylation level is lower than the healthy seedlings, the DNA methylation can be elevated to a higher level when the phytoplasma was eliminated. Therefore, the reaerach on the relationship between phytoplasma and paulownia DNA changes may be beneficial fox paulownia witches’broom occurred elucidation of the molecular mechanism and establishment of the prevention methodsPaulownia tomentosa × Paulownia fortune, Paulownia fortune, and Paulownia tomentosa were treated by different concentrations of Dimethyl sulfate (Dimethyl sulphonate, DMS), observing the changes of seedlings morphology,at the same time, the nested PCR was used to detect phytoplasma, the SSR and AFLP were used to analysis the changes of the DNA base sequences. the summarize as follows:(1)Suitable concentration of DMS treatment turned the disease seedlings into healthy seedling in morphology:after 3h treatment,15 mg · L-1DMS bud still appearing symptoms of witches ’broom seedlings in morphology, while the time up to 5 h or the concentration more than 15 mg · L-1 the seedling are showing the health morphology, the phytoplasmas can not be detected but 15 mg · L-1DMS/5 hby nested PCR.(2)The DNA base sequence of Paulownia witches’broom seedlings, healthy seedlings and DMS treatment seedlings was not found changes on the SSR level, on the level of SSR molecular markers, amplified by45 pairs of primers, the results showed that Paulownia’ DNA restriction sites did not found changes by DMS treatment(3)The DNA amplified site of Paulownia witches’ broom seedlings, healthy seedlings and DMS treatment seedlings was not found changes on the AFLP level, on the level of AFLP molecular markers, amplified by 96 pairs of primers, the results showed that Paulownia’DNA base sequence did not found changes by DMS treatment... |
