Different part of seedlings(cotyledonary node,cotyledon,hypocotyl) by in vitro culture, young embryo, anther and adventitious root were used as explants in this experiment to study the influence of different hormone on adventitious bud and adventitious roots formation, a regeneration system of explants in vitro culture was established; The contents and composition of fatty acid compounds of eight Eucommia seed samples from four different provinces were extracted by Soxhlet extraction and detected by Gas chromatography (GC), meanwhile, the unsaponifiables in the seed of eight Eucommia ulmoides Oliv was detected by GC/MS. The composition of fatty acids and sterols exist differences in these seeds from different areas. This study benefit of the further study of in vitro culture and rapid propagation of Eucommia ulmoides Oliv.The major findings are summarized as follows:1. On solid, liquid MS medium supplemented with lmg/L TDZ, different parts( cotyledonary node, hypocotyl,cotyledon) of 2-week-old Seedlings were used plant to induce adventitious buds, no adventitious buds were induced on liquid medium, on solid medium inductivity was:100%,65.9%,39.0%;2. On medium supplemented with different concentrations of 6-BA,2,4-D, TDZ and 6-BA mixed with 2,4-D, TDZ, NAA, GA3, entire young embryo during different periods(65,73,81 days after pollen dispersal) were used to plant on young embryo culture. The results was:young embryo of 73 days after pollen dispersal were the best explant; the best medium of induction adventitious buds and embryogenic callus were MS+2mg/L 6-BA and MS+0.5mg/L 2,4-D; the inductivity was91% and 100%.The result is similar to the mature embryo. The optimum medium for embryoid was MS+0.5mg/L 2,4-D and inductivity was 8%. This result has not been reported before.3. Cotyledon,hypocotyl and cotyledonary node of 2-week-old Seedlings and cotyledon of embryo induced adventitious roots on MS midum with 0.2mg/L NAA, inductivity was 93%,62%,37%,98%.The adventitious roots were used as material to induc adventitious buds, the effect of different hormones were TDZ> 6-BA> ZT> KT;The optimum medium for adventitious buds was MS+TDZ 2mg/L+NAA 0.3 mg/L; inductivity was 65%.4. The optimum medium for the regenerated buds rooted was MS+ 0.2mg/LNAA; the rooting rate was 50%.5. Flower bud with slightly stretched branches cultured in 25℃ or stored in low temperature (4℃) after 60 days, anther could develop to mononuclear period which could be used as explants for in vitro culture; Anthers stored at 4℃ low temperature as pretreatment after 7 days had more distinct stimulative effect on callus induction, in the opposite shorter duration (2 days) of pretreatment showed inhibition occurs; The optimum medium for anthers was 2mg/L 6-BA+0.5 mg/L NAA,1mg/L 6-BA+lmg/L NAA+2mg/L TDZ; inductivity was 95.9%,100%.6. Fatty acid composition and content between seeds of different provinces of have a little difference. A total of 12 kinds of fatty acids have been detected, and there are 10 kinds of same fatty acids in all measured the samples, the highest content is Linolenic acid (55.50%~61.57%). Difference also exists in composition and content of sterol between different seeds from these provinces; Total sterol content of seeds from Jiang xi Jiu jiang is the highest, the content could reach 8.27 g/kg and the lowest is seed from Jiang xi Jiu lian shan which content was 8.27 g/kg.
