In order to evaluate their genetic structure, the genetic diversity in Jiangxi(Poyang Lake)、Hubei(Liangzi Lake) and Jiangsu(Taihu Lake) population of Erythroculter ilishaeformis was analyzed based on RAPD method. Results showed that twenty polymorphic primers were selected among 30 arbitrary primers. These 20 polymorphic primers could not only amplify the common DNA bands of three population, but also amplify particular alleles in the fish.119 amplified fragments were detected ranging from 0.2kb to 1.7kb. The number of RAPD polymorphic sites in Poyang Lake population was 61, accounting for 51.26%. The number of RAPD polymorphic sites in Liangzi Lake population was 50, accounting for 42.02%. The number in Taihu Lake population was 67, accounting for 56.30%. The Shannon genetic diversity index of Poyang, Liangzi and Taihu Lake was 0.4292,0.3828 and 0.4431, respectively. The genetic similarity between Poyang and Liangzi Lake was highest among the three population (0.8055), followed by that between Poyang and Taihu Lake (0.8016), and the genetic similarity between Liangzi and Taihu Lake (0.7852) was the lowest. Conclusions:These results could provide the basis for selecting parents of improved variety selection.
