Crucian(Carassius auratus)is an important economically freshwater fish in Northern Asia which can adapt the extremely high alkalinity. The soda lake Dali Nor is extremely high alkalinity water in which living saline-alkaline tolerance crucian population。China has a lot of alkalinity area, most of them has not been exploited. The Dali Nor population multiply in Dali alkalinity lake is good experimental material to study alkaline-tolerant genes. Using NGS(Next Generation Sequencing)we can explore functional gene and analysis DEG(Differential Expressed Genes). Our study get high-throughput RNA-Seq data from gill and kidney of alkalinity water of Dali Nor and fresh water of Ganggeng Nor to understand the adaptation of crucian against extreme water environmental stress.Our study found that 3433 and 3420 differentially expressed genes in the gill and kidney, respectively with FDR(false discoveryrate) ≤ 0.01, FC(fold-change) ≥ 2, FC(fold-change) ≤-2. Result shows that known functional categories of genes and signaling pathway significantly enriched, they associated with stress response and extreme environment adaptation, the functional categories are response to stimulus, oxidoreductase activity, transporter activity, transferase activity. Our study identified significantly DEGs of aminotransferases, carbonic anhydrases, encoding important modulators on stress adaptation and tolerance, including, heat shock proteins, superoxide dismutase, glutathione S-transferases and so on.This study showed details and information of gene expression changes in extreme environmental stress in crucian,provide a solid foundation of fish breeding in saline environment for further study.
