Trichokonins(TKS) is a variety of antibacterial peptides isolated from T.pseudokoningii SMF2, which have a broad spectrum of bactericidal activity. Ailanthus altissima can absorb dust and harmful gases, purify environment, eliminate noise and increase the humidity of air,which be used as street trees. Its special smell has strong ward off effect to many kinds of pests,so it can reduce the dangers of pests on other trees. Ailanthus can absorb heavy metals or other harmful substances to speed up the recovery rate of repeal and serious polluted industrial land.But the seedling of Ailanthus is disease sensitivity and die as it is too weak. The Ailanthus seedlings was as a target plant and the fungus diseases as target fungi to research the TKS in this study. Which was to research the antibacterial activity and mechanism of TKS. Results showed that,(1) 5 kinds of funguses were pathogenic bacteria for Ailanthus seedling in the 8 kinds of isolated funguses.The methods of isolate single spore, molecular biology, microscopic observation and Koch’s postulation were used in this study to identification the eight kinds of isolated funguses. They are named Fusarium equiseti, F. soiani, Acremonium implicatum, Alternaria tenuissima, Pestalopsis clavispora, Chaetomium globosum, Engyodontium album and Paraphoma radicina. F. soiani is the main pathogenic funguses, followed by F. equiseti and C. Globosum, then are A. Tenuissima and P. clavispora.(2) Inhibit the spore germination and the growth of hypha is the bacteriostatic mode of TKS.The mycelial growth and spore germination methods were used to research the inhibitory effect of TKS to hypha growth and spore germination on pathogenic funguses. In this study, the five kinds of pathogenic funguses were as the target, the Carbendazim,Difenoconazole, Matrine and Downy mildew like were as the positive control. Results showed that,TKS can inhibit the hypha growth and spore germination. The medicament concentration more high, the inhibition effect more significant. The bacteriostatic effect of TKS is poor than Carbendazim and Matrine, is similar to Difenoconazole, they are strong than Downy mildew like. It can control the disease caused by F. Soiani, C. Globosum, A. Tenuissima and P.Clavispora effectively, the inhibition effect on F. Equiseti is poor. The inhibited effect on spore germination was superior to hyphae growth.(3) The TKS can induce the resistance of plants.The TKS can promote the growth of Ailanthus seedling in a certain concentrationrange. It can increase the growth indexes, such as plant height, total root length, fresh weight and dry weight. It can affect the physiological indexes of plants, such as increase the content of soluble protein, proline and soluble sugar, promote the synthesis of chlorophyll a, inhibit the synthesis of chlorophyll b and the the content of MDA, O2-and H2O2, so it can reduce the damage to plants. What’s more, it can promote the enzyme activity of SOD, POD, PPO and PAL to inspire the resistance of plants.(4) The TKS could make the form of pathogen hyphae and spores changed.The TKS could cause the spore plasmolysis, hypha deformation and internode shorter when it in a high concentrations.(5) Inhibiting cellulase activity is one of the mechanisms for TKS.TKS can inhibit the cellulase’s activity, such as mycelium carboxymethyl cellulose enzyme and 1,2 beta glucan enzyme which is one of the mechanisms for TKS.(6) The TKS has triple efficacy of growth agents, biological agents and chemical agents.The TKS has the characteristics of growth regulators, as it can promote plant growth in low concentration and inhibit the growth of plants in a high concentration. It is easy to be absorbed by the plant, safety and low toxicity. What’s more its efficacy play slowly but persistent. So it has the characteristics of biological agents. As it can inhibit the growth of pathogenic bacteria or die them. So it has a dual characteristic of biological agents and chemical agents. |
