Rice stripe disease is reported to cause severe losses in rice fields in temperate regions of East Asian. To date, the interactions study among RSV protein and rice protein or among RSV proteins has been not reported systematically. So, there is the important value on understanding the pathogenic mechanism of RSV by studying their interactions. In this work, we studied the interaction among the proteins coded by- RSV and- rice, or between the proteins coded by RSV using yeast two-hybird system(YTHS) and bimolecular fluorescence complementation(Bi FC), the results were indicated as follow: firstly, a protein coded by RSV can interact with many rice proteins. For instance, many proteins were simultaneously screened against Rd Rp as bait, such as phosphoribulokinase, chloroplastic-like(PPKase), photosystem I reaction center subunit II chloroplastic-like(PSAD2), mitogen-activated protein kinase(MAPK), and NADH kinase-like(NADK3), et al. Secondly, a rice protein can simultaneously interact with many RSV proteins, such as ribulose-1, 5-bisphosphate carboxylase/oxygenase large subunit(RBCL) can interact with almost all of proteins coded by RSV, NADH dehydrogenase can interact with RSV P2 and MP. Lastly, many rice proteins can interact with the RSV protein, such as abscisic acid receptor PYL8-like and translation initiation factor e IF4 A can interact with RSV P2, poly(A) binding protein and light harvesting complex mesophyll7 can interact with RSV P4. Based on above the studies, the interaction relation was further verified by Bi FC in N. benthamiana.In addition, the results by YTHS indicated that the self-interaction of Rd Rp-1,-2,-3,-5, P2, Pc2 C, CP and MP was confirmed; the two-way interaction of P2 and Rd Rp-1,-2, P3 and Rd Rp-2, P2 and Rd Rp-5, P2 and Pc2 C was confirmed; one-way interaction of Pc2 C and Rd Rp-1, P2 and Rd Rp-4, P2 and P4 was also confirmed.Based on above all interaction results, we draw a network map of protein-protein interactions, and RSV P2 protein was displayed to be an important protein, which can interact with many RSV proteins.
