| Porcine arthritis is a tissue inflammation caused by one or more factors, and bacteria are main pathogens. According to reasearches, The main pathogens of porcine arthritis included streptococcus suis(S.suis) and haemophilus parasuis(HPS), Staphylococcus and so on. Because most of pig farms are influenced by arthritis, there are severe economic losses in the swine industry in our country. It reported that S.suis and HPS can cause other clinical symptoms as well. With the variation of inheritance, the two pathogens showed multi-drug resistance and measures to the clinical prevention and control have been complicated.In order to understand the epidemiological characteristics of main pathogens of porcine arthritis in East China, the multiplex(2) PCR to detect capsular polysaccharides(CPS) gene of streptococcus suis serotype 2(S.suis2) and 16 s rRNA gene of HPS was developed first. Using the method to test 120 strains of bacteria isolated from samples suspected porcine arthritis in East China, there are 14 strains of S.suis2 and 12 strains of HPS. Due to 21 strains of S.suis were detected in 120 samples, S.suis2 were considered the most popular in S.suis. To research the virulence factor distribution type of S.suis isolates by series of multiplex PCR established furtherly, it proved a low-toxic type of +cps2J-/+mrp-ef-sly-gdh-/+gadph-/+orf2. Comparing the cps2 J gene of S.suis2 isolates with four domestic strains and two foreign strains of S.suis2, the identification homology was 94%-99%. Meanwhile, genetic evolutionary tree of cps2 J gene showed that the S.suis2 were euroky in East China. The restriction fragment length polymorphism(RFLP) of tbpA gene of four strains of HPS isolates showed all of the isolates are DBE type which is the most popular genotype. The drug sensitive test of S.suis isolates and HPS isolates showed multi-drug resistance, and the drugs of penicillins and cephalosporins were sensitive to both of S.suis and HPS. Therefore, the both of drugs can be used to treat porcine arthritis.The research provided a rapid and acurate method for detecting porcine arthritis in pig farms. The detected reasults accumulated useful informations for etiology research of porcine arthritis in East China. The preliminary analysis of main virulence factors of pathogens offered a reference for the development of vaccine in the future. And the susceptibility test provided a guidance for developing the measures of prophylaxis and treatment of porcine arthritis. |
