Background: With the rapid development of domestic Life science, the demand for high quality of experimental animals is increased. Currently Chinese situation, the scarcity of SPF level of experimental animals has become one of the reasons affecting the development of biological technology in China.Objective:In order to solve this situation, establish the SPF Japanese big ear rabbit animal population by the biopurification technology. By this way it can increase the supply of the SPF experimental animals and so that it also do well in the research. For domestic SPF rabbits breeding population and promotion provide the reference.Methods: First of all, using the method of Cesarean delivery to the Japanese big ear rabbit of ordinary level to purify, and then having a caesarean birth. Use the SPF Japanese big ear rabbit to instead of lactation, measuring the Ph in whole blood, and combining with some indexes or physiology, biochemistry, clinical and so on. So that it can establish the SPF Japanese big ear rabbit animal population,and supply the large number of demand.Results: 328 rabbit babies gained from the germfree caesarean section. The livability of the 286 survivals after weaning is 87.2%, which is high survival rate. 8 groups, 16 males and 80 females were selected as production groups. The SPF barrier system ran smoothly in this one year and gained 1980 SPF Japanese white laboratorial rabbits with high quality.The comparation experiment proved barrier system didn’ t have obvious harm to SPF Japanese white laboratorial rabbit, moreover, the growth-development performance of Japanese white laboratorial rabbits in the barrier system was better than Clean Japanese white laboratorial rabbit, but this difference has no statistics meaning. There are significant difference between SPF Japanese white laboratorial rabbits and Clean Japanese white laboratorial rabbits on blood physiological and biochemical indexes, ten organ coefficients.In the process of the biopurification technology, deal the 40 pregnant rabbits with the caesarean section, giving birth to the little rabbit number is 328,Out of the breastfeeding is 286.The survival rate is 87.20%,the number is remarkable. Selecting 16 male rabbits and 80 female rabbits divided into 8 groups as the producting population. The SPF barrier system runs stably, with out pollution lasts one year, obtained the SPF Japanese big ear rabbits 1980.Through the contrast experiment shows that, barrier system can keep the level of the experimental animals as the SPF. By the way growth was slightly better than the clean level Japanese big ear rabbit, but the difference was not significant. But some indicators is significant difference in the blood biochemic and the immunology.By using strict operation specifications of laparotomy and foster rabbit, we can get more ablactational rate than artificial nursing. It is effective to maintain SPF system in the barrier system under the strict SOP system. Base on biological elimination technology andstrict SOP system, can get large high quality SPF animal in short time, which also provide new ideas for experimental animals production-oriented enterprises.
