| Leptin is a protein hormone secreted by fat cells that can reduce energy consumption, and increase energy consumption. Leptin play a role in physiology through signal transduction pathway by combine with its receptor.In this experiment the metabolic disorder mice induced by CLA for the model, adopt the immunohistochemical methods to study of liver metabolism related gene expression changes after leptin interference.CLA is added in the feed, We successful induction of metabolic disorders mouse which have hyperglycemia, hyperlipidemia, fatty liver index and so on.Experimental is divided into three groups: linoleic acid(LA) group as control group,experimental group linoleic acid group have experimental control group CLA+saline(CLA+S) group and experimental group CLA+leptin(CLA+L) group.Kunming mice born 21 d after weaning, feeding added LA and CLA rat chow, feeding after 42 d, LA groups don’t do processing, the mice of CLA underwent subcutaneous implantation of an micro-osmotic pump containing either saline or leptin at a dose of 1mg/ml for 2 weeks.Then took the liver to do immunohistochemical test. According to the results, CLA group mice compared with LA group, the expression of Akt, p-Akt and FOXO1 reduced which related with sugar metabolism, and in the CLA+L group, the expression of Akt, p- Akt and FOXO1 quantity increase. Accordingly, CLA can restrain the express of these several gene, made insulin signaling pathway can’t smooth conduction, thus, the phenomenon of the high blood sugar and insulin resistance appeared. CLA+L group compared with CLA+S group, the expression of these genes rise,explain that leptin can regulate the expression of these genes to improve the sugar metabolic disorders in the body. Leptin can contribute to the transfer of insulin signal downstream singnal and expression to regulating glucose metabolism.Leptin through conbine with its receptors, pathways downstream of p-STAT3 of CLA+L group significantly increased compared with CLA+S group expression quantity(p < 0.05), leptin signaling pathway signal transduction enhance. The key enzyme of G- 6- Pase play an important role in sugar dysplasia aspects, CLA+S group compared with LA group, the expression was significantly decreased(p < 0.05), sugar dysplasia activity decreased;CLA+L group compared with CLA+S group, the expression amount significantly increased(p < 0.05), sugar dysplasia increased activity. Lipid metabolism, CLA+S group compared with LA group, CLA+S group the expression of SRB1 decreased significantly(p < 0.05), the expression of CD36 significantly increased(p < 0.05).Shows that CLA can reduce the expression of SRB1 and at the same time can increase the expression of CD36. Speculation SRB1 activity increasing in the reverse cholesterol transport, CD36 express quantity increases and increase the intake of long chain fatty acids, accumulation in the liver and cause fatty liver.CLA+L group compared with the CLA+S group, the expression of SRB1 significantly higher(p < 0.05), lower the expression of CD36.SRB1 in the reverse cholesterol transport activity is abate, CD36 mediates the long chain fatty acids in the diet reduce. |
