Distant cross is an important way of creating new species.The similar relationship between bamboo and rice in the evolutionary tree,add the same dyeing base,provide the possibility for distant hybridization.In order to get hybrid seeds from the experiments of hybridization between bamboo and rice,which can provide new genetic resources for genetic breeding of bamboo.In this experiment,we generated 16 combinations of hybrid tests.the results found only in the Lei bamboo and rice hybrid combinations,harvested 170 hybrid seeds.after sowing,seedling,survival 130. By flow cytometry,found a plant genome size was located between bamboo and rice,we called it suspected hybrid plant.Through morphological observation,the plant type of suspected hybrid rice including plant height,leaf length,leaf width and stem were between parental bamboo and rice;Stem with segment was obvious,this feature was same to bamboo,while leaf type like rice;Hairy leaf surface was different from parental bamboo and rice.Using rice SSR primer on the suspected hybrid was labeled verification,found the primer were poor transferability and polymorphism in Lei bamboo,then SSR primer of lei bamboo was developed.We get 132678 cDNA sequences available from Transcriptome sequencing.and found 18356 simple sequence repeat(SSR) loci comprising repeats of one or two or more nucleotides in 15600 cDNA sequences. The rate of cDNA sequences which containing SSR loci in total is 11.75%.SSRs consisted of 49.6% mononucleotide repeats,24.6% trinucleotide repeats,24.2% dinucleotide repeats.The most frequent SSR motif in cDNA sequences was was(A/T)n and(CCG/CGG)n was the second one. 299 SSR loci were selected for marker development,out of those 30 primer pairs resulted in PCR products of the expected size and SSR motif and were developed and used to assess the transferability and polymorphism of the bamboo collection consisting of 21 differment species,3species of phyllostachys,DNA genetic variation of 14 variation types of Ph.violascens,Transferability of EST-SSR was the best at the level of intra-genus,Polymorphisms were evident at the level of sympodial versus monopodial bamboo species.The new developed SSR primer used to the suspected hybrid,and found the plant had some same strips with Lei bamboo,illustrate the validity of hybrid.
