| This article based on RNA- Seq technology,Sequenced and made analysis of the sheep blastocyst and hatch-blastocyst transcriptome, To obtained a series of datas,analysis of the dates and got the the main results of the research are as follows:(1) Sequenced the sheep blastocyst and hatch-blastocyst Transcriptome through Illumina/Hiseq2000 platform,made a series of work include filtration,restructuring and annotation,ect. Obtained 47664034 and47821016 Clean reads respectively,Compared with the reference genome,the proportion of reads reached71.48% and 72.90% respectively.(2) The number of transcripts in blastocyst and hatch-blastocyst transcriptome were 19318 and 18288,The number of transcripts that the coverage reached90%-100%. Predicted 2879 novel transcripts in blastocyst,1277 novel transcripts in hatched-blastocyst.(3) A total of 7700 differentially expressed genes,Blastocyst-vs-hatched blastocyst 3129 gene expression level up regulated,4571 gene expression level down regulated.(4) There are four types variable junction site in Blastocyst and hatched- blastocyst,Exon skipping,Intron retention,Alternative 5’ splice site,Alternative 3’ splice site.The number of them are 29117 and 11332;6864 and 5698;14899 and 8190;16321 and 9431.(5) The number of SNP site selected from blastocyst and hatched- blastocyst are 97620 and 67471,There are two types of substitution:transition and transversion,most is transition.There are 5461 Extend Gene in blastocyst and 5904 ExtendGene in hatched- blastocyst.This Experiment involved basal date analysis and advanced analysis in the transcriptome of sheep embryos.Provided abundant dates for the next step research about the transcriptome of sheep embryos.At the same time, the select and analysis of Differentially expressed genes will provide basic theory for gene regulation happened in the process of sheep embryos.The prediction of novel transcripts,the selection of variable junction site and SNP site Greatly widened information of the sheep transcriptome,provided date reference to sheep embryo development gene regulation especially the process of blastocyst and embryo before implantation. |
