This article aims to explore the effects on growth performance and biochemical indexes in serum of holstein bull calf afteri njecting umbilical cord mesenchymal stemc ells,the experiment one collected umbilical cord tissue from a healthy holstein newborn calf, purified and cultivated umbilical cord Mesenchymal Stem Cells(UC-MSCs) in vitro by the method of organization to adherent and Pancreatic enzyme digestion.and observed its biological characteristics,cell growth curve of the first, the fifth and the tenth generations was drawed. Osteogenic differentiation capacity induced was detected by histochemical staining(Alizarin Bordeaux staining) in vitro and adipogenic differentiation capacity induced is detected by Oil Red O staining method in vitro.Results show that the cell generation of classical culture still keep a good shape, proliferation ability to keep consistent with 1 to 5 generations. Osteogenic cells induced in vitro for 21 d was proved that there has been induced osteogenic differentiation capacity by von Kossa stained calcification nodules of skeleton cells in cell culture plate,in the meanwhile,adipogenic cells induced in vitro for 14 d was proved induced adipogenic differentiation capacity by fat droplet in cell culture plate.It showed that bovine umbilical cord mesenchymal stem cells can culture and proliferate in vitro,and having characteristics of differentiation capacity of mesenchymal stem cells.28 Holstein calves were selected with conditions of be born for one month and similar weight, then they were divided into control and experimental groups. Calves of experimental group were injected in jugular vein with the Umbilical Cord Mesenchymal Stem Cells which diluted to 8 mL( 3 × 105 cells /per kg), the control group was injected with normal saline. The test lasted for 154 d. It was the same of cattle diet formula, nutrition level, and management conditions between experimental group and control group in test duration from bull calf 1 months to six months,before every injection, calves were tested some indexes sunch as weight, body height, body length, chest circumference, pipe circumference and waist wide Angle on an empty stomach. Adopted blood 5 ml by Jugular vein and stored in- 20 ° C, 3500 r/min, the centrifugal 15 min,serum Ca, P, TP, ALB, BUN, TG, CHO, ALP, LDH, CK, GH, LEP andSS, T4, T3, IGF-1, VEGF, bFGF, TGF-α, BMP-7, IgA, IgM, IgG and IFN-γ, IL- 1, IL 2and IL 6and MDA, SOD contents were entrusted Beijing huaying biological co., LTD.Results: After injection of UC-MSCs, the average weight of experimental group was significantly higher in the 4, 5 and 6 months of age the control group(P<0.05); average daily gain of experimental group rather significant exceeded control group in 3,4 months old(P<0.05); while at 6 months of age, which difference was very significant(P<0.01);Body size of the test group at the peak of the growth and development of male calves were higher, and the difference was significant(P<0.05); Serum P content in the experimental group in 3, 4, and 6 months of age was significantly higher than the control group(P<0.05), serum Ca content in experimental group at 5, and 6 months of age is significantly higher than the control group(P<0.05); When 4 months of age, the test group of ALP, LDH, CK in serum were significantly higher(P<0.05),,the test group were significantly lower than the control group of BUN in serum(P<0.05); during 5 months of age, the test group of Alb, ALP levels in serum were significantly higher(P<0.05); 6 months of age, the test group of TP levels in serum were significantly higher(P<0.05), and between the two groups CK levels in serum were different significantly(P<0.01); When four months old, the experimental group of SS levels in serum was significantly lower than the control group(P<0.05); the experimental group T3, T4 levels in serum difference was very significant when six months of age(P<0.01); while at 6 months of age, which difference was very significant(P<0.01).Serum insulin-like growth factor(IGF-1) level of experimental group was showed significant higher than the control group in 4 months of age(P<0.05);During 3 months, the two groups of serum vascular endothelial growth factor(VEGF), transforming growth factor(TGF-α) showed significant difference(P<0.05); 6 months of age, the experimental group of VEGF, b FGF, TGF-αlevels in serum were significantly higher(P<0.05).While 2,3,4,6 months of age, the control group and the experimental group BMP-7 level in serum appeared significantly different(P<0.05);The IgA level of experimental group in serum was very significantly higher in 4 months than control group(P<0.01), while at the age of 6 months, which difference was significant(P<0.05); the two groups of IgG in serum showed very significant difference in 2、5 months(P<0.01), and the difference was significant in 6 months(P<0.05); During 3、4 months, Serum IL-2 level of experimental group was showed significant lower than the control group(P<0.05); experimental group Serum MDA level in 2, 3, 4, 5, 6-month-old were significantly lower than the control group(P<0.01); At the age of 5 months, the experimental SOD levels in serum were significantly higher(P<0.05), while in 6 months, the difference was very significant(P<0.01).Conclusions:Thus, injecting umbilical cord mesenchymal stem cells improved significantly the Ca, P, TP,ALB, ALP, LDH of the bull calf serum, reduced the level of BUN, CK in serum, and stabilized the inter environment,and promoted the synthesis and deposition of bull calf body protein and bones, in the while, umbilical cord mesenchymal stem cells significantly improved the VEGF, TGF-α, bFGF, BMP 7 levels in serum, reduced the hormone level of GH, SS, LEP in serum, improved the level of T4, T3, these growth factors and hormone levels in serum were closely related to the growth and development of bull calf, influenced indirectly its weight and body measurements, increased the development speed of holstein bull calf, and through improved immune factors such as level of IgA, IgG, reduced the level of serum MDA and increased SOD activity can enhance the body’s ability to clear free radicals, reduced inflammatory factors IFN-γ, IL-1, IL-2,IL-6 concentration in serum, reduced internal inflammation, thereby enhancing male calf immunity, promoting the healthy growth and development of bull calf will affect its growth performance.
