The main purpose of this study was to egg yolk substitute beta cyclodextrin inclusion compound Cholesterol Cholesterol-loaded Cyclodextrins (CLC) shock to cold and freezing and thawing pig sperm quality parameters and the influence of the mitochondrial membrane potential. This experiment USES the quality of landrace boar semen, different treatment, divided into three groups, respectively, test: freeze-thaw, cold shock and normal temperature preservation processing, detection of sperm vitality and mitochondrial membrane potential, the results are as follows:1. Add CLC set of sperm motility characteristics:cold shock and freeze-thaw sperm group average linear velocity (VSL), the curve of average velocity (VCL), average path velocity (VAP) were higher than in did not add group, add 1.5 mg/ml CLC experimental effect is remarkable2. Add such as concentration of CLC in different incubation time found in the experiment on the influence of sperm vitality, sperm motility highest 15 minutes of incubation test group.3. With coomassie brilliant blue staining method to assess, cold shock and freeze-thaw pig sperm acrosome integrity, according to the results of 1.5 mg/ml CLC cold shock group,1.5 mg/ml CLC (196℃) group freeze-thaw acrosome intact rate is better than the other group.4. By adding different concentrations of CLC experimental application of mitochondrial membrane potential detection kit (JC-1) sperm mitochondrial membrane potential, the results showed that adding 1.5 mg/ml cold shock treatment group and add 1.5 mg/ml of freeze-thaw treatment group sperm mitochondrial membrane potential is higher than other groups.In a word, in the process of pig sperm cryopreservation add CLC on sperm viability will have a positive impact. |
