Mycelial Chemical Constituents And Antioxidant In Ophiocordyceps Lanpingensis Populations

In this study,13kinds of chemical constituents in mycelium of14different populations of Ophiocordyceps lanpingensis Yu et Chen were detected and analyzed; and two groups (20strains of each group) strains were chosen to test the activities of polysaccharide antioxidant and flavonoids antioxidant, respectively. The goal was to provide a scientific basis for development and utilization of O. lanpingensis.The13kinds of chemical constituents of O. lanpingensis populations were detected by colorimetry and HPLC, respectively. Polysaccharides, mannitol, flavonoids, uracil, uridine,2’-deoxyuridine, inosine, guanosine, adenine, thymidine, adenosine and2’-deoxyadenosine were all detecTab. in mycelium of O. lanpingensis populations, while the cordycepin had not been detected. The variation patterns of these12kinds of chemical constituents in different populations varied at different degrees. The2’-deoxyuridine had maximum degree of dispersion, which the average variation coefficient among populations was105.291%, and204.171%among individuals in each population. The maximum variation coefficient was175.883%in Shi-deng population (LPSD), and the minimum variation coefficient was45.825%in Ying-pan population (LPYP). Flavonoids had the minimum degree of dispersion, which average variation coefficient was25.909%among populations, and33.766%among individuals. The maximum variation coefficient was40.409%in East-Mountain population(LPTDS), and the minimum variation coefficient was15.696%in Ju-dian population(LPLIJ).The analysis of variance indicated that the differences of the content of chemical constituents in different populations is significant. The difference of adenosine was largest and the differentiation between populations was most significant. The adenosine average content of each population was higher than the quality standard (100μg/g) of O. sinensis in Chinese Pharmacopoeia. The104Snow-Mountain population had the highest content of adenosine which was1093.007μg/g. But the difference between it and Lian-he vilage and Jian-chu Mountain populations was not significant with, while the difference between it and others was significant (P<0.05). The stain LP104XS031had the highest content of adenosine(1529.661μg/g), and its content of uridine, guanosine, flavonoid were also relatively higherwhichwere1838.967μg/g,1796.949μg/g,2156.249μg/g, respectively.The correlation analysis showed that there were different degrees of correlation between most components. Most constituents had low linear correlation, only the correlation between uridine, adenosine and guanosine were high linear correlated, and the correlation coefficients between guanosine and adenosine, guanosine and uridine, adenosine and uridine were0.894,0.858and0.811, respectively. The nested analysis indicated that76.077%of the average variation of mycelium chemical constituents was resided among individuals, and23.924%was remained among populations. It was showed that the mycelium chemical constituents had differentiation among populations in O. lanpingensis. The variation of variance were9.94%-35.84%. And adenosine, uridine, guanosine and mannitol had larger differentiation among populations.The antioxidant activity of O. lanpingensis mycelium was detected in the present study. The polysaccharide and flavonoid extracted from mycelium sampled both had antioxidant activity. The more sample concentration was increased,the more antioxidant activity was enhanced. The polysaccharide and flavonoid antioxidant activities in most strains of O. lanpingensis were higher than that of O.sinensis. The strain LPJCS15of O. lanpingensis was the best one in the aspect of polysaccharide antioxidant activity. The IC50value of scavenging DPPH free radical and hydroxyl free radical were1.556mg/mL,1.161mg/mL, respectively. And the absorption value of total antioxidant capacity was0.089when the sample concentration was2.5mg/mL. The strain LP104XS031of O. lanpingensis was the best one in the aspect of flavonoids antioxidant activity. The1C50value of scavenging DPPH free radical and hydroxyl free radical were0.967mg/mL,3.743mg/mL, respectively. And the absorption value of total antioxidant capacity was0.236when the sample concentration was2.5mg/mL.In summary, the variation pattern of mycelium chemical constituents in different populations of O. lanpingensis was revealed among populations and individuals. The variation of the chemical constituents mainly remained among individual strains. The polysaccharide and flavonoids of O. lanpingensis mycelium both had the antioxidant activity. Most strains of O. lanpingensis had higher antioxidant activity than O. sinensis. The strains LPJCS15and LP104XS031had higher antioxidant activities, but also had good quality comprehensive index of active ingredient. It was suggested that both strains could be used for large-scale fermentative production.
