MicroRNAs (miRNAs) is a class of small RNA molecules (~22nt) and plays an important role in biological function. The inhibition of translation or induce degradation of protein-coding mRNAs contain complementary sequences to mRNAs and plays an important role in many specific biological processes, thus affecting all aspects of the biological activities of life. This study identified bovine specific-miRNAs by bovine mammary epithelial cells,while detected microRNA in milk whey and compared with microRNA in goat’s milk whey. The test is divided into two parts:1. miRNA expression profiling is affected by many factors, including tissue types, developmental stages, health or disease, and a variety of nutrients, and so on. This experiment prepared bovine mammary epithelial cell line as experimental material in Ruminant Nutrition Lab of Animal Chinese Academy of Agricultural Sciences, to explore miRNA expression and identification. We extracted RNA from a library in the BGI companies by Illumina/Solexa1×36bp sequence data and analyzed RNA by RT-PCR. This experiment identified388known miRNAs and38newmiRNAs members which some of them havetissue-specific portionor tissue distribution limitation.2. The experiments determined11specific microRNAs in early mammary epithelial cell model, based on the raw milk whey from two batches (before sterilization) extracted directly microRNAs.We determined the contentof specific microRNAsthrough the reference gene and we found the expression of miR-2284-k、miR-2284-x、miR-2904-1、miR-3432-1and miR-2887-1show high abundance based on the relative gene and suggested species specificity.
