Eperythrozoonosis is caused by Mycoplasma suis. It can cause fever, hemolysis, jaundice and other symptoms in clinic. It causes a serious risks and hazards in swine industry. In recent years, there has been a lot of cases about M.suis mixed infection s with pleuropneumoniae, porcine circovirus, pasteurellosis, et al. It is difficult to cur e this kind of disease. In order to reduce the incidence and prevalence of this diseas e in clinic and the losses caused by the disease to farmers. It is important to preven t from the disease. The immunity of M.suis was preliminarily studied in this paper. The white oil and propolis adjuvant vaccine of M.suis were prepared and it has som e value to prevent Eperythrozoon suis.The study is divided into three parts,1):The M.suis antigen preserved in our lab was detected by Giemsa staining and molecular biology. The primers were designed based on AJ504999. A fragment of781bp was amplified and it’s similarity to a know n sequence of M. suis(accession numbers AJ504999) was99.4%. But fragment was n ot obtained in the control group.2):Preparation and detection of inactivated vaccine of M.suis. The condition of inactivation of M.suis was explored including the time a nd temperature of inactivating, the concentration of inactivator formaldehyde. The mo st suitable time and temperature were24h,37℃respectively,and the concentration of formaldehydewas0.4%. The vaccine of M.suis were obtained by adding white oil a nd propolis adjuvant with emulsification method. The formulations, uniformity and sta bility, viscosity and other physical and chemical properties of adjuvant vaccine were detected. The safety of vaccine were tested in mice. The result indicated the vaccine were safe.3):The study of animal immunization experiments and immunogenicity.T he mice and pigs were immunized with the prepared vaccine. The index of spleen a nd pancreas were detected in mice. Compared with the control group the indexes of immune groups were higher. It indicated that the the vaccine could enhanced the im munity of mice. No symptoms of Eperythrozoonosis appeared after the immunized mi ce were infected with M.suis. The antibody levels of immunized pigs in serum were measured by ELISA. The antibody levels of immunized groups were increased. The r esults of Lymphocyte transformation test showed lymphocyte transformation test result s showed that the OD value of immunized groups were increased. There were differe nces between immunized groups and control group. In addition, the SOD and MDA i n immune serum was detected. The results of SOD showed that the capacity of antio xidant and scavenging free radical metabolic were not changed.The results of MDA h ad no change.It showed that the health of bodies were normal. In addition, the ALT and CREA were measured and indicated it had no effect on liver and kidney.
