The different areas forsythia in 4 provinces Shanxi, Henan, Shaanxi, Hebei were used in this study, based on the 18 different habitats’forsythia and adulterants forsythia Viridissima, Syringa oblate, Forsythia mandshurica Uyeki was analyzed to genetic diversity by using ISSR molecular marker technology. The main results are showed as follows:1. The experiments respectively using the method of CTAB method, high salt and low pH method and SDS method to filter and optimize the extraction of forsythia suspensa’s DNA. The results showed that, the extraction of high salt and low pH method of DNA got good purity, protein and polysaccharide removing completely.2. A total of 34 primers were screened from 100 ISSR primers which published by Columbia University when studied the genetic diversity of two populations of Radix Astragali. The results showed that 16 primers among them were polymorphism.3. Analysis of genetic diversity in different kinds of Forsythia suspensa, among them SXLC’s Ne, h, I was highest. It shows that SXLC were abundant genetic diversity and Allele maximum effect, and HNLX’s Ne、h、I was the lowest.Analysis on genetic diversity of different kinds of ShanXi, HeNan, HeBei, ShaanXi, the results showed that the level of genetic diversity in ShanXi forsythia (Na:1.7420; Ne:1.3010;h:0.1916; 7:0.3052)was the lowest in the four provinces. Therefore, its genetic information and genetic diversity was the least.But the average of four provinces were 1.8145,1.3013,0.1940,0.3125, respectively. This indicates that the four provinces have larger genetic information and genetic diversity.4. Analysis on genetic differentiation of ShanXi, HeNan, HeBei, ShaanXi from different provinces. The results showed that, the coefficient of gene differentiation among ShanXi’s populations(Ht:0.2030, Hs:0.1657) was more higher than HeNan (Ht:0.1863,Hs:0.1528), HeBei(Ht:0.1843, Hs:0.1722),ShaanXi(Ht:0.1897, Hs:0.1713). Similarly, The genetic differentiation were as of ShanXi’s populations(Gst:0.1835, Nm:2.2249), HeNan(Gst:0.1799, Nm 2.2799), HeBei(Gst:0.0656, Nm:7.1166), ShaanXi(Gst:0.0969, Nm:4.6611). In summary, the degree of genetic differentiation among populations: ShanXi> HeNan> ShaanXi> HeBei, HeBei and ShaanXi have stronger gene flow.5. Using UPGMA to 18 origin of forsythia and its adulterants Admiralty flowers, Northeast forsythia, lilac by NTSYS analysis. Different regions of the forsythia and its adulterants were divided into 4 groups in 0.70. Among them,18 origin of forsythia belongged the first group and the rest three adulterants are respectively a group. It showed that different origin of Forsythia suspensa in a certain extent and location is a certain contact.Generally different provinces in the same province of Forsythia Fructus Forsythiae clustered firstly.
