Objective:TFA was used as object in order to study the anti-inflammatory and immune effects and its mechanism of TFA in vitro.Methods:The LPS-induced and normal RAW264.7 cells were used to build models in vitro, experiment was divided into blank groups, model groups, TFA groups (low, medium and high). Doses of TFA were determined by cell toxicity test. The level of NO was detected by Griess kit. The levels of PGE2, IL-6, IL-1β, IFN-γ, TNF-a were detected by ELISA kits. The expressions of iNOS and COX-2 mRNA were detected by RT-PCR method. The COX-2 and iNOS proteins and phosphorylation levels of ERK, JNK, p38 were determined by Western blot method.Results:1. The levels of NO and PGE2 in normal RAW264.7cells were raised by TFA low and medium group. The levels of NO, PGE2, IL-6, IL-1β, IFN-γ, TNF-α in LPS-induced RAW264.7cells were reduced by TFA groups.2. The expression levels of iNOS and COX-2mRNA in nomal cells were raised by TFA low group. The expression level of iNOS and COX-2mRNA in LPS-induced cells was reduced by TFA groups, the effect was in a dose-dependent relationship.3. The levels of iNOS, COX-2 proteins, and phosphorylation of p38, JNK protein in nomal RAW264.7cells were raised by TFA low dose group. The levels of iNOS, COX-2 protein, and phosphorylation of p38, JNK protein were reduced by TFA groups, and the effect was in a dose-dependent relationship.Conclusion:The study showed that TFA regulated the levels of NO, PGE2, IL-6, IL-1β, IFN-γ, TNF-α by regulating the levels of iNOS and COX-2 mRNA and protein, and the phosphorylations of p38 and JNK, and played anti-inflammatory and immnue two-way regulatory effect in vitro.
