| Based on two kinds of wild Zoysia varieties from different habitats(Zoysia japonica Steud. cv. No.43and Zoysia japonica Steud. cv. No.133), the physiological and biochemical characteristics under drought and low temperature stress was studies, include the change of REL, RWC, Fv/Fm and primary metabolites.36kinds of metabolites were measured from Zoysia under drought and low temperature stress by GC-MS technique, including10kinds of organic acid,14kinds of amino acids,7kinds of soluble sugar,2kinds of fatty acids,1sugar alcohol,1polyol and1amine. The result showed that:1. The results showed that RWC and Fv/Fm were significantly decreased in the two Zoysia japonica varieties under cold and drought stress, and the decline under low temperature stress was significantly than dought stress. At the same time, the RWC and Fv/Fm in WZG43showed a significantly smaller decline than WZG133.2. The relative electric leakage dramatically accumulated in the two Zoysia japonica varieties under cold and drought stress, and the accumulated under low temperature stress was significantly than dought stress. It’s showed that low temperature stress on two kinds of Zoysia japonica than drought stress caused more serious loss, more to change the cell membrane permeability and damage the photosynthetic system more.3. The metabolites of WZG43and WZG133were measured by GC-MS technology, and a total of identified36kinds of metabolites were identified, including10kinds of organic acid:propanoic acid, ethanedioic acid, phosphoric acid, propanedioic acid, butanedioic acid, butanedioic acid, trihydroxybutyric acid, ribonic acid, gluconic acid, benzoic acid;14kinds of amino acids:alanine, valine, isoleucine, glycine, serine, threonine, aspartic acid, proline, glutamic acid, phenylalanine, asparagine, glutamine, lysine, tyrosine;7kinds of soluble sugar:fructose, glucose, mannose, arabinofuranose, sedoheptulose, sucrose, trehalose, two kinds of fatty acid:hexadecanoic acid, octadecanoic acid; moreover including myo-inositol, glycerol, ethanamin.4. Except for benzoic acid,35kinds metabolites showed a different accumulation in Zoysia under drought and low temperature stress. Zoysia accumulated these metabolites under drought and low temperature stress in order to osmotic adjustment, maintenance of cell turgor pressure and rebuilding a new balance of metabolic homeostasis. On the other hand, these metabolites are primary metabolites that can repair the damage which caused by drought and low temperature stress. |
