Adipose tissue is important to both energy storage and metabolism regulations,and is is crucial for the meat. Intramuscular fat is a major indicator of the tenderness,juiciness and flavor of beef. Preadipocytes can be induced to adipocytes bydifferentiation.Proliferation and differentiation of preadipocytes affect both lipiddeposition and intramuscular fat, and then influence the meat.The studies have shown that TNNT1,HBA,HBB,MB,MYLPF,MYL1,PGAM2,MYL6B,HSPB1,TPI,MSRA,TNNI2,TNNT3,TCAP,TXN,PSMA1,GSTM1,COX6B1,VDAC1,GSTP1genes have differential expressions in beef cattlewith different fat deposition abilities. First, we cultured bovine preadipocytes, andinduced to be adipocytes. We detected the expression of these genes in preadipocyteby qRT-PCR both before and after the inducing. The lipid deposition abilities werealso measured by Oil Red O staining. Results indicated that the mRNA and proteinexpression of PSMA1, HBB, COX6B1, MYLPF, TPI, PGAM2, HBA, TCAP, GSTP1,MB, TXN, GSTM1, MYL6B, MSRA, HSPB1, TNNT1were increased afterpreadipocyte differentiation but expression of VDAC1and MYL1were decreased.There was no expression of TNNI2, TNNT3. The lipid deposition was also increasedobviously after differentiation. It suggested that these genes may play potential rolesin preadipocyte differentiation and lipid deposition.There was the most differential expression of PSMA1gene after thedifferentiation of preadipocytes, so we detected the the effect of PSMA1gene onbovine preadipocyte differentiation and lipid deposition. Silencing of PSMA1geneby Lipofectamine2000, the lipid deposition abilities were also measured by Oil RedO staining and triglycerides, the ability of differentiation were tested by differential expression of PPARγ, C/EBPα, LPL and Pref-1.The result showed the lowexpression of PSMA1could inhibit the formation and accumulation of lipid droplets,and decrease the number of triglycerides and the mRNA expression of PPARγ,C/EBPα, LPL.The results provided the theoretical basis of bovine fat deposition,thus, taked the gene as a molecular marker of beef quality traits and providestheoretical basis for early breeding and selection.
