| ABSTRACT In this study, the cDNA sequence of LFY and API were cloned from the blade and petal of Nelumbo nucifera’Hongxiamantian’ by RACE and RT-PCR。1Cloning and analysis of the full length cDNA of LFY from the blade of lotus by RACE and RT-PCRWith the basis of known sequence, obtained the5’and3’end sequence by RACE. And the full length cDNA of LFY is1494bp, it contains a largest ORF that is1173bp encoding390amino acids (Mw=65110.0), a5’untranslated region of69bp and a3’flanking sequence of252bp.The deduced amino acid sequence of LFY was compared with the sequences of other higher plants such as Solanum lycopersicum, Dendranthema lavandulifolium, Cucumis melo,ilium brownii var. viridulum,and so on.Conserved domain analysis showed that this protein belongs to the FLO_LFY gene family.2Cloning and analysis of splicing sequences of API from the petal of lotus by RACE and RT-PCRWith the basis of known sequence, obtained the3’end sequence by RACE.And the partial sequences of API is788bp, it encodes226amino acids (Mw=25831.9), and contains a3’flanking sequence of110bp.The deduced amino acid sequence of API was compared with the sequences of other higher plants such as Eschscholtzia Californica, Nandina domestica, Petunia hybrida,Cabomba caroliniana,and so on.Conserved domain analysis showed that this protein belongs to the MADS-box gene family. |
