| Streptomyces roseoflavus Men-myco-93-63is an actinomyces agent isolated from thesoil inhihiling potato scab and its metabolite have evident antagonistic effects against manypathogens, the strain such as Verticillium dahliae, Sphaerotheca fuliginea, Botrytis cinereaPers. et al.., the solid fermentation and liquid fermentation of the strain technology hasriped. To make the better use of strain in the production practice，the Men-myco-93-63nutrient tablets applied in the soil has been developed, and we detected its characteristicsand the control effect against cotton verticillium wilt. The main results were listed asfollows:The carbon and nitrogen source has been screened by single factor and double factorsvariance analysis. And the nutrient tablets of Men-myco-93-63was prepared, thecharacteristics were detected. Then we optimized the storage means of the tablets related towater content, temperature, and vacuum. The results showed is that the rato of kieselguhrand fermentation liquor of Men-myco-93-63is2:1. The optimum carbon and nitrogensource are15%of sucrose and peanut meal respectively. The adhesive is corn starch orPEG6000. The prepration process of tablet is as follows: kieselguhr and fermentationliquor of Men-myco-93-63were incorporated thoroughly, then the carbon and nitrogensource and adhesive were added and incorporated thoroughly. Dried in low temperature,smashed in the granulator, tableted in pressure machine. The processed nutrient tablet hasthe characteristics. The facade is clean and smooth, color uniformity, edges neatly, meetsthe standard of weight difference, and no cleavage fracture. After the tablet storage, withwater content increasing, the content of actinomycetes first increased then declined, andreached a maximum at2%moisture content; lower temperatures can prolong the storage;and encouraged the growth of bacteria in the air. The best storage solutions was watercontent2%, at4℃and vacuum processing. Actinomycetes content which can stillachieve4×105cfu/g in120days, and contamination rate was less than1%.In field the effect of the nutrient tablets of Men-myco-93-63against cotton verticilliumwilt was tested, the number of soil actinomycetes, the yield of cotton, state of an illness ofverticillium wilt also were tested, to evaluate the influence of the nutrient tablets. The results showed is that1.0g nutrient tablets of Men-myco-93-63was better than othertreatment in the content variation of actinomycetes, the effect against cotton verticilliumwilt, advance of the cotton yield. After50d, amount of colonization of Men-myco-93-63still reached up to4.9×105cfu/g, the effect against cotton verticillium wilt and the yield oflint cotton were88.66%and2245.50kg/hm2respectively, the yield increase36%. |
