Nucleic acid preparation as a new biological products,has great potential forprevention and treatment diseases such as infectious diseases, genetic diseases, cancer andthe improving of growth and reproductivity of animals. According to the internationalstandards, nucleic acid preparation need to be strict safety evaluation, before large scaleproduction and application. Whether the preparation can integrated into the hostchromosome DNA is one of the most concern of the people.Early research in our laboratory, A nucleic acid preparation,which can effectivelyimprove silk chicken egg production performance has been develpmented. We used Qiagencompany production of plasmid kit preparation for injection of nucleic acid preparation.Through the establishment of a complete set of safety evaluation, and according to thesampling plan design of cycle, then shot the application kit to extract the nucleic acidpreparation of Rhode Island Red chicken genome of all the related organizations, includingmuscle (injection organization), intestinal, ovaries, liver, stomach, heart, fallopian tubes,spleen, kidney nine groups.Three repeated sampling three chickens. Sampling time,respectively0,3days,7days, Later, the sampling every seven days. And design thecorresponding primers for PCR amplification, observation records of the plasmid DNA. Apreliminary analysis of the preparation indicates that the preparation effectual workingtime is about21days. Obviously it is short, for this reason on the basis of the originalpreparation The SV40large T antigen expression structure andSV40replicationoriginwere added into the preparation. These made, the effectual working time of thepreparationwas extended to49days.This study also tested distribution in different tissues and persistent time of thepreparation by PCR detection method, and preliminary analyzed on its security. Get thefollowing results: In the muscle tissue of the nucleic acid preparation sustainable existenceto49days,56days has completely undetectable; For the chicken heart,liver, stomach andbowel tissue with edible value, In the third days can detect trace, in the seventh day hascompletely undetectable, which shows that these organizations are safe to eat after7days;For the fallopian tubes and ovaries of the reproductive organs, namely can’t detect its existence in the zeroth day, presumably eggs are edible. Of course, these are theoreticalspeculation, still not finalized its safety. Any nucleic acid drug product before large-scaleapplication development requires strict biological safety assessment.In short, the project development for nucleic acid preparation of egg, although thereare a lot of work remains to be further developed, has good development potential.Therefore, it is suggested that further increase investment in order to develop in-depth andmeticulous research, listed as soon as possible for the products and services to the societyto lay a solid foundation.
