From the the late20th century to now, set off a wave of stem cell research in the world, theunique characteristics of the proliferation and differentiation of stem cells has attracted more and moreattention.With the rapid development of increasingly sophisticated and various theories of stem cellrelated technologies will bring a major revolution in the traditional medical field. Stem cells have theability of self-renewal and pluripotency, they are very consistent with the principle of preservation ofanimal genetic resources, preservation and utilization and they can be used in it.In this research, weisolated PMSCs of Chinese Luxi cows,identified the biological characteristics and the differentiationpotency of them.Draw the following results:1. Established the cultural system of bovine PMSCs. The suitable culture condition for PMSCs wasL-DMEM+10%FBS.2. Established the bovine PMSCs cell line, which expressed the markers of mesenchymal stem cells,having strong proliferation and Multilineage Differentiation Potential,The PMSCs were subcultured toas many as passage65, and were cryopreserved for203vials in all.3. We identified bovine pancreas mesenchymal stem cells morphology and surface markers,It isconsistent with the characteristics of PMSCs.4. We identified PMSCs by RT‐PCR.The results showed that PMSCs expressed the MSCs markers of CD29, CD44、CD73、CD90、CD106、CD166、Vimentin,Insulin and nestin while did not express endothelial cells specific marker CD34and CD45.5. Bovine pancreatic stem cells can be differentiating into osteoblasts and adipocytes.
