Peanut derives from South America. It is one of the four largest oil crops in the world.Peanut is one of the most competitive agricultural exports agricultural commodities of Chinain international. With the rapid development of peanut industry, aflatoxin pollution problemshave become increasingly prominent, production safety and pollution-free peanut products isimminent. It is an effective way to use tissue culture and genetic engineering breedingtechniques to cultivate anti-aflatoxin peanut varieties. This article studied several aspect ofembryos lobular of FengHua3, LuHua11, HaiHua1and HuaYu19from the aspectsof adjusting hormone ratio, and aimed to screen appropriate adventitious buds differentiationmedium. To select the best peanut varieties-differentiation medium combination and studygenetic transformation of resistance genes. The results are as follows:(1) PnLOX2gene which was connected to the prokaryotic vector was successfullyconnected to the PRI101-AN, and transferred to the Agrobacterium-EHA105.(2) Hormone adjust the medium, screening the optimal bud induction differentiationmedium of Fenghua3, Luhua11, Haihua1and Huayu19. The most suitable differentiationmedium hormone ratio for Fenghua3is6-BA:NAA=6:0.2; the optimal hormone ratio ofLuhua11is6-BA:NAA=6:0.8; the optimal hormone ratio for Haihua1and Huayu19is6-BA:NAA=4:0.8. The most suitable rooting medium for regeneration of Fenghua3isMS0+2mg/L NAA.(3) The selection pressure for Fenghua3is adding100mg/L Km and400mg/L Cef todifferentiation medium; The rooting selection pressure for Fenghua3is adding13mg/L Kmto rooting medium medium.(4) Genetic transformation of peanut and obtain transgenic plants, the transgenicplants obtained by trace PCR detection showed positive reaction, then concluded that thePnLOX2gene was transferred into the peanut.
