The method using the hybrid complementary effects develop the cotton yield, quality and resistance has got more and more attention as multi-objective cotton breeding requirement (such as high production, high quality and resistanceetc.) was raised, and utilization of the advantages of cotton cross-fertilize has become an important channel to further improve cotton output and quality.Traditional heterosis breeding, consisting of male sterile line, maintainer line and recovery line three substeps, was a time-consuming and difficult, which trans breeds male sterile line to screen out the advantages combination of high cotton combining ability and to obtain recovery line possessing excellent quality, and the production trait was reduced as this method could not completely maintain the original combination of heterosis in trans-breeding process. Another method, adopting artificial emasculationmanner to produce cotton hybrid seed, was high-cost and also limited by the flowering time, the scale of hybrid cotton production was thereby restrictedtremendously. Therefore, controllable trangenosis ofmale sterility genes into cotton through biological approaches has momentous economic values.In this paper, infertility gene vehicle p3PBATBsystem was applied, which was constructed by Professor Xiao from China Agricultural University and has the ability of chemical induced recoverable fertility.Xinluzao42somatic embryogenesis and plant regeneration system were established firstly, and then the35regenerated plants could be obtained through genetic transformation of the p3PBATBvehcle system.The experimental results illustrate that (1) callus were effectively produced on the mediums with hormone regime of four types and embryonic callus were appeared only on medium with IBA1.0mg·L-1+KT0.5mg·L-1or2,4-D0.1mg·L-1+KT0.1mg·L-1hormones combination. Embryonic callus of the cultivar Xinluzao42were effectively initiated in46d on medium with IBA1.0mg·L-1+KT0.5mg·L-1the medium with2,4-D0.1mg·L-1+KT0.1mg·L-1must be transferred to medium of MSBP for subculture. Embryonic callus in the MSBF medium covered with filter paper differentiate into embryo and concurrent plant regeneration. It took about140d to obtain the regeneration plants The successful regeneration system established in the Cultivar Xinluzao42.(2) Regenerated plants were obtained by transforming infertility gene vehicle p3PBATBsystem, which has the ability of chemical induced recoverable fertility.Finally,33positive regenerated plants were gained after anti-PPT screening and PCR detection.
