Thinopyrum bessarabicum Love,2n=2x=14, JJ or EbEb shows resistance to multiple diseases and good salinity tolerance, which represents an important genetic resource for wheat improvement. Stressed with250mmol/L NaCl, the multiple translocation and addition line TJ02shows good salinity tolerance for several years, which not only contains a pair of chromosome6J but also contains a pair of segmental translocation involving2JS. To allocate the tolerance genes, series of alien chromosome lines including DA6J, DS6J (6A), DtA6JL, DtA6JS and T6AS-6AL-2JS were recovered from hybrids F2-F3between TJ02and Chinese Spring and characterized by both molecular and cytogenetic markers.40markers specific for6J which included28on6JS and12on6JL were identified and the breakpoint of T6AS-6AL-2JS was mapped at C-6AL4-0.55. Tested with250mmol/L NaCl, both TJ02and CS-Thinopyrum bessarabicum amphiploid showed better tolerance that CS. The salt harm index and Na+/K+of lines DA6J, DS6J(6A), DtA6JL, DtA6JS and T6AS-6AL-2JS decreased in a different degree than CS indicating tolerance of TJ02might be controlled by multiple genes which might be contained in both chromosome6J and T6AS-6AL-2JS.124collinear markers specific for seven chromsomes of Thinopyrum bessarabicum were identified, which included11on1J,25on2J,12on3J,14on4J,9on5J,28on6J and13on7J. Characterized with these markers and FISH/GISH, three reciprocally translocated chromosomes added in wheat were confirmed which included T6JS-2JL, T3JS-4JL and T3JS·7JS, the other new additions involving intact chromosomes2J,3J and7J including DA2J, DA7J and MA3J were then recovered from the hybrids of CS and CS-Thinopyrum bessarabicum amphiploid. Thus, a set of CS-Thinopyrum bessarabicum alien chromosome lines involving seven intact chromosomes of Thinopyrum bessarabicum has been developed, which will be very useful for mapping, transferring and utilizing beneficial genes of Thinopyrum bessarabicum. |
