Ammopiptanthus mongolicus belongs to the Leguminosae plants, is a national secondary protection plant, is unique to the desert region of northwest China super xerophytic evergreen broad-leaved shrub, has important ecological significance and economic value.This article applies the tissue culture technology, which used Ammopiptanthus mongolicus young stem, root and annual stem segments as explants to research the callus induction, axillary buds and clump buds induction technology, and the proliferation, single bud elongation and rooting induction technology, establishes the regeneration plant system. The results of the study are as follows:This study used the young stem segments as explants, consulting the induction rate, proliferation coefficient, growth status and so on, the medium of axillary buds induction is: MS+1.5 (mg/L) 6-BA+0.1(mg/L)KT+0.5(mg/L) IBA; the medium of axillary buds proliferation is:MS+0.5 (mg/L) 6-BA+0.1 (mg/L) KT0.1(mg/L)IAA+0.2(mg/L)IBA; the medium of single buds elongation is:MS+1.5 (mg/L) 6-BA+0.1 (mg/L) KT +0.5(mg/L)IBA. Used the root tips as explant, the medium of clump buds induction is:B5 +1.5 (mg/L) 6-BA+0.1 (mg/L NAA); the medium of clump buds proliferation induction is:MS+1.0 (mg/L) 6-BA+0.1 (mg/L NAA); and adding 0.5 mg/L GA3 makes that, the bud seedlings can grow up to 3.45 cm. Used the annual stem segments as explant, the medium of axillary buds induction is:MS+1.5(mg/L) 6-BA+0.5 (mg/L)NAA+0.5IBA (mg/L).The experiment uesd different explants, the research results show that the best explant is the root tip. According to the rooting rate, the average root length,the rooting coefficient, and other factors, the best medium of rooting induction is:1/3+0.2(mg/L) IBA+0.5 (mg/L)NAA. |
