| One of the important ways to improve the efficiency of crop breeding work is combined molecular breeding and conventional breeding. In this paper, confectionary sunflower inbred lines for test materials were studied the relationship between setting rate faceplate and other agronomic traits, and markered the materials with SSR mark-ers:The result as follows:(1) Plant seed rate Setting rate with number of leaves per plant, plant height, stem diameter, single-disc grain weight, single-disc grains, faceplate diameter was posi-tively correlated, in which the largest correlation coefficient was with single-dish grain weight. Plant seed rate, grain weight of single disc, single-disc grains, grain weight of these four traits cumulative contribution reaches 80.202%> 80%. estab-lished multiple linear regression analysis plant seed rate (Xi), single disc grain weight (Y), single disc grains (X2), kernel weight (X3), the regression equation Y= 23.42+ 0.546Xi+0.01X2+0.17X3,and to set up a best-fit regression equation:Y= 0.498Xi+ 16.643. Y= 738.528+6.568X2+0.005X22, Y=9.24+0.123X3-0.001X32when the threshold is 10 sunflower seed rate as the parameter for the test material is divided into four categories group.(2) Assuming DNA template from four-leaf stage of confectionary ers’leaves for experimental materials, this study used the single factor test and or-thogonal design for optimizing six SSR-PCR reaction system factors (10×PCR buf-ferã€Mg2+ã€dNTPsã€, primersã€Taq DNA polymerase and different concentrations of Mg2+ã€dNTPsã€Taq DNA polymerase and DNA template on SSR-PCR reaction system. The result shows that the range of the influence of six factors on the reaction system is Mg2+> Taq DNA polymerase (primer)> DNA template>dNTPs. And this study estab-lished confectionary sunflower SSR-PCR reaction system,10×PCR buffer 0.2mmol/L, Mg2+ 2.0mmol/L, dNTPs 1.8mmol/L, Taq DNA polymerase 0.2u, DNA 50ng,primer primer 1.5mmol/L.(3) 50 pairs of primers were obtained 317 polymorphic loci,per primer pair 6.34 polymorphic loci, the genetic similarity coefficient varies as follows:0.5268 to 1, with an average of 0.6963(4) After markers 102 sunflower inbred lines using molecular marker technique of SSR cluster analysis to these materials,when the genetic similarity coefficient was 0.62 the tested varieties were classified as four types of groups, in the four types of groups,there were 5 species in the I group theses species accounted for 4.9% of the tested varieties, the group II includes two subgroups there were 17 species in total that accounted for 16.7% of the tested varieties, the groupâ…¢ includes two subgroups there were 21 species in total that accounted for 20.6% of the tested varieties, the group IV includes three subgroups there were 59species in total that accounted for 57.8% of the tested varieties. |
