Early studies showed that retrotransposons atrl participated in spur variation of apple. In order to study the expression activity of atrl, this experiment used quantitative PCR to research the expression activity of atrl by different stresses with atrl-LTR,which provided a reference for creation= mutant by retrotransposons atr1 in the future.Using young apple trees as test materials to do siress treatments of S A,2,4-D, ABA and wounding, of which water treatment was used as the control. RNA was extracted and reverse transcribed into cDNA after digested by DNase and primers were designed to do RT-PCR experiment The test results showed that retrotransposons atrl expressed in control sample and each treatment in different degrees; The fluorescence quantitative analysis showed that ABA, wounding and 2,4-D could imcrease the expression activity of atrl with GAPDH as internal control; The analysis in different time after 2,4-D treatment showed that atrl transcriptional activity presented high-low trend, and the expression activity reached the peak after 9 hours treatment.The germplasm resource of jujube was abundant in Anhui. Jujube witches’broom caused by phytoplasma have been an important obstacle that threated the local jujube industry in recent years. This research had analyzed 16S rDNA sequence characteristics of jujube witches’broom in Anhui; The content of cytokinin and auxin in jujube shoots between healthy plants and those infected with phytoplasma were compared; Part of the fragment of ipt gene was cloned.1. Study the sequence characteristic of 16S rDNA from jujube witches’broom in Anhui Province. Using the DNA extracted from Fanchang long jujube’s healthy and plexiform symptoms leaves as template, We cloned 16S rDNA sequence of phytoplasma from jujube witches’broom, and gained a fragment strain JWB-FJP1 with 1248bp in length. The sequence homology analysis showed JWB-FJP1 shared the highest identity of 99.92% with Chongqing jujube witches’broom phytoplasma strain JWB-Xch(JQ675716) and shared the lowest identity of 67.06% with Japan jujube witches’broom phytoplasma strain JWB-Hebei(GU184186), and it was classified as a member of 16Sr V-B group. The result indicated that phytoplasma shows different biological characteristics in different hosts.This research provides a theoretical basis for fast identification and classification of jujube witches’broom phytoplasma in Anhui local jujube species.2. The determination of the content of cytokinin and auxin in jujube shoots between healthy plants and in plants infected with phytoplasma. The contents of cytokinin and auxin in jujube shoots between healthy plants and phytoplasma-infected jujube plants in May and July were detected by ELISA, and the result showed that the content of cytokinin phytoplasma-infected was higher than that in healthy plants.3. To clone the fragment of isopentenyl transferase gene. Using the DNA extracted from Fanchang long jujube’s healthy leaves as the template and the homologous part of ipt gene accessed in NCBI to design primers, we gained a fragment of 311bp in length after PCR amplification, recovery, clone and sequencing. It was found that this fragment shared the identity of 73% with mulberry and shared the identity of 69% with glycine max.
