Study On Culture System In Vitro Of Wild Chive "Jinxiu"

The technology of plant tissue cultivation is mainly adopted in the study. The wild chive "Jinxiu" from Guangxi is used as the experimental materials. The study is carried out by the choice and disinfection of explants, primary cultivation, bud-propagation, strong seedling, root-breeding, exercising seedling, and transplanting. The aim is to find the best solution of cultivation of vitro culture system for wild chive "Jinxiu". The most suitable culture medium and hormone proportion, the best concentration of organic additives, and the optimal sucrose concentration, etc., are included in the best solution of cultivation. The results show that:The key procedure of establishing primary cultivation system is choosing the best explants and combinations of the exogenous hormone. The best explants are non-blossomed flower disk and fully-blossomed flower buds. The differentiation rate of callus and bud of non-blossomed disk are 100%. The best hormone combination of callus differentiation for blossomed flower buds is MS+6-BA2.0mg/L+NAA1.0mg/L +KT0.5mg/L, its rate is 80.48%. The best hormone combination of bud differentiation for blossomed flower buds is MS+6-BA2.0mg/L+NAA1.0mg/L, its rate is 48.56%.Among the sterilization treatments of blossomed flower buds, the effect of double factors is superior to the single factor. The best sterilization combination is 75% ethanol30s+0.1%HgCl212min, and the contamination rate is 23.04% without influencing the differentiation of explants. In the same sterilization treatment and different blossoming states, the contamination rate of flower buds is list in ascending order:non-blossomed, semi-blossomed, and fully-blossomed.The best medium and combined hormone of leaf propagation is MS+6-BA 2.0mg/L+NAA1.0mg/L, the multiple of which is 2.93. The optimal sucrose concentration is 30g/L, the multiple of which is 2.95. The optimal concentration of coconut milk is 20%, whose multiple is 3.83.The medium,1/2MS, is better than MS medium on root-breeding of wild chive "Jinxiu".The rate of root-breeding of 1/2MS medium is 100%, and the average amount of root is 6.38, and the average diameter of root is 0.97mm, and the average length of root is 4.24cm, and the average height of plant is 13.58cm. IAA performs better than NAA and IB A on the induction of root-breeding, the best concentration of IAA is 0.5mg/L, and the rate of root-breeding is 100%, the average amount 8.33, the average diameter 0.94mm, the average length 3.34cm, and the average height of plant is 12.13cm. Among the double factors treatment, 1/2MS+IAA0.5mg/L+PP3330.3mg/L is best for root-breeding, the rate of root-breeding is 100%, and the average amount of root is 10.7, the average diameter 1.27mm, the average length 3.11cm, and the average height of plant is 11.91cm.Either single garden soil or be matched with other culture substrates in some rate can be used for transplanting. The best culture substrate is two portions of import peat soil+one portion of perlite among the treatments of culture substrate. The single garden soil should be best if considering economic factor. On this condition, the survival rate is 100%, and the average height of plant is 12.74cm, and the average length of root is 2.25cm. 属性不符...
