As a traditional Chinese medicine in China, aloe vera, due to the unique active ingredients, are widely used in medicine, food, cosmetic,etc. But the research is few as a botanical fungicide in the prevention and control of agricultural disease. This experiment studied the antibacterial mechanism of aloe acetone extracts against tomato late disease pathogen, the results are as follows:1. Made by acetone as solvent, the average extraction yield of aloe vera extract was 67.3%.2. The minimum inhibitory concentration of aloe vera extract was 1.67 mg/mL.3. Compared with the control group, by the results of SEM knowing that after tomato disease pathogen with aloe vera extract, it happened to the surface morphology of the sag, distortion, perforation cracking phenomena, and bacteria; By TEM as you can see, after the aloe extract solution treatment, tomato late blight fungus cell wall thinning first, then the irregular shape of wool cloth with soft nap, finally cytoplasmic extravasation due to cell wall broken seriously and appear empty capsule.4. The result of effect of the aloe vera extract on the cell membrance of tomato late blight fungus showed that conductivity has increasing trend after treated. Due to damage to cell membrane, the influence on permeability caused internal electrolyte exosmosis. Further study found that the leakage of macromolecular substances of tomato late blight fungus is serious, and discovered by PI tag showed leakage of large molecules exist in the DNA.5. For EB fluorescence probe, to study effect of the aloe vera extract on DNA of tomato late blight fungus, the result showed that aloe extract of antimicrobial substances can similar to EB with tomato late disease pathogen DNA and DNA binding, so that the fluorescence intensity changes, the conformation of DNA that aloe extract can destroy bacteria, eventually killing bacteria.
