This research use transgenic technology to salt tolerance model plant Euphrates poplar and transgenic model plant Arabidopsis as the research object. To build and use have been Euphrates poplar genome into bacterial chromosome library, by agrobacterium tumefaciens mediated, using the model plant arabidopsis thaliana as receptors, through the inflorescence infiltration method, will carry a random Euphrates poplar genetic infect into arabidopsis thaliana, larger pieces for populus euphratica arabidopsis thaliana genome into library. And through the resistance training, PCR technology method of screening positive plant, observe the screening of leaf number, leaf, plant type and the abnormal shape, flowering of differences on plant such as morphology, significant difference of strain and the performance to carry on the key observation records, identification, the author analyzes the reasons of the phenotypic differences between, provide the basis for study of p. euphratica art gene screening. The results of the study are as follows:(1) Of this study is to activate agrobacterium strains 491, received 166 positive transformation strain, arabidopsis transformation success rate was 33.8%, because the successful large fragments into arabidopsis transgenic strains.(2) The study of the communist party of China on the 522 plant DNA extraction and PCR identification, including 384 positive plants, positive detection rate of 73.56%, confirmed that most of them into plant positive for genetically modified plants.(3) In this study for large positive plant phenotypic analysis, found a total of four phenotypic changes of mutants, after further screening, confirmed that the mutation phenotype can stable genetic gm strains.(4) This research focus on 45 alb7 eight offspring strains of flowering time and bolting time, leaf shape, Indus, leaf area, petiole length, leaf width, leaf and so on carries on the observation records, and its data by SPSS 19.0 significant difference analysis. Results showed that the mutant and wild type in flowering time and bolting time, leaf shape, Indus, leaf area, petiole and veins are significant difference, it is for the mutant.This study will carry a random Euphrates poplar genetic infect into arabidopsis thaliana, larger pieces because the successful transformation of arabidopsis thaliana genome strain, and obtain stable phenotype mutation of arabidopsis thaliana. This study shows that the system can be successfully applied, because the woody plants and herbaceous plant arabidopsis thaliana research, provide the basis for populus euphratica art gene identification analysis.
