Study on the natural color silk is attracted many people’s eyes in recently. This article focuses on the green cocoon silkworm variety because of its rich in flavone pigments and when silk reeling its color is not easy to fade, these reasons become the major research chooses. But the flavonoid pigments on green cocoon silkworm in vivo metabolic process、transport mechanism and the genes regulation mechanism、cocoon color genetic and many other aspects are still incompleteness.This paper firstly provides a breeding method of natural green cocoon silkworm variety which bred by ourselves, study on its each tissue, larvae pupa and cocoon of silkworm flavonoids content in fifth instar, respectively, compared with the white cocoon variety. Research also on green cocoon related gene UGT86 and the flavones synthase gene BmFNS I in silkworm midgut, fat body and the silk gland, the fluorescent quantitative PCR technology is used to analysis. Discussion on the flavones pigment differences between tissues of silkworm and the green cocoon formation mechanism, provides a new reference for the formation of green cocoon, also provides meager strength for the silkworm breeding of green cocoon variety. This paper has investigated and compared some characters of two varieties, studied the absorption of ultraviolet of green cocoon and white cocoon, and the UV protective property of cocoon silk cloth. To provide evidence for the green cocoon as new functional materials, applying for the textile, cosmetic, medical and other fields. The main results are as follows:1. This paper provides a method for cultivating natural green silkworm variety.According to the mechanism of genetic regulation of green cocoon and cocoon fluorescence color of sex linked gene Fx with green cocoon genes. Using the hybridization, generation of directional breeding methods, while maintaining the current good economic characters of white silkworm varieties, the improvement and breeding combined with GcGcFxFx genes of green cocoon variety of silkworm.2. The flavonoids content of white cocoon and green cocoon in layer and pupa.Flavonoids content in green cocoon layer showed highly significant difference. Although the flavonoids content in green cocoon pupa is also higher than white cocoon pupa, did not show significant difference.3. The flavonoids content of each tissue in two silkworm varieties.Compared with midgut, blood, silk gland and fat body of two silkworm varieties, the flavonoids content in each tissue has the difference in different days, midgut content is relatively higher than other tissues.4. The relative expression of UGT86 and BmFNS I.Comparison of relative expression of two genes in the midgut, silk gland and fat body, the expression of BmFNS I show a high expression. Different tissues expressions are different.5. Investigation and analysis traits of white cocoon and green cocoon.From the age of time, cocoon quality performance and silk performance to investigate two varieties. The green cocoon variety age than white cocoon variety is short about 60 hours. Whatever the whole cocoon weight in both male and female, cocoon weight and cocoon layer ratio, the green cocoon show inferiority than the white cocoon silkworm varieties. The reason is probably with the full age through time is shortened. Silk achievements in many aspects of green cocoon varieties are lower than those of the control variety, but can be considered as the natural color silk quilt material and pharmaceutical use as biomaterial.6. The cocoon layer UV reflective degree and made of cloth the UV protective property and ultraviolet transmittance.Green cocoon is significantly lower than the white cocoon layer in short wavelength of UV reflective degree, maybe that the ability of green cocoon to absorb short UV stronger than the white cocoon shell. The test cloth values of UPF and transmittance UVA and UVB, get the conclusion that green cocoon variety obvious better than white cocoon variety, we conclude that the UV protective property of green cocoon variety showed a big advantage.
