| In this study, RBP4 and BMP7 genes which are candidate genes about porcine reproductive performance were selected, its SNPs (single nucleotide polymorphisms) were detected and associated with litter size in two Alien species pigs (Large white and landrace pig) and two Anhui superior native pig breeds(Huoshou Black Pig and Wannan Black Pig). Primers were designed according to Sus scrofa’ BMP7 (Number:NC010459) and RBP4 (Number:NC010456) gene which were published in NCBI, polymerase chain reaction-single-strand conformation polymorphism (PCR-SSCP) and sequencing were used to detected the SNPs and the GLM model analysis in SPSS program were used to detected the associations between the genetic variation and sows’litter size, to find out the the effective genetic markers of reproduction traits in Anhui native pigs,, and provide reference of molecular biology for the study of germplasm characteristics and MAS in Anhui native pig breeds. The study show that:(1) the SNPs analysis of RBP4 and BMP7 genesThere was no mutation in exon 1,5,6 in RBP4 gene in these four populations; Moreover there was no mutation in exon 2,3,7 in BMP7 gene in these four populations.(2) genotype frequencies and gene frequencies of the SNP sites of pig RBP4 and BMP7 genesThe results showed that an A→G mutation in intron 3 at 604 bp,3 genotypes (AA, AB and BB) were found in RBP4 gene in Large White Pig, Landrace Pig and Wannan Black Pig, and no mutation was detected in Huoshou Black Pig. The dominant allele is allele A in this three pig breeds. The AB was the dominant genotype in Lardrace Pig and Wannan Black pig, the genotypic frequency was 49%,43.24% respectively.The AA was the dominant genotype in landrace pig, the genotypic frequency was 46.23%. The polymorphism information content (PIC) was 0.3463,0.3589,0.3598 respectively in Landrace, Large White and Wannan Black populations, they were all in average polymorphism situation. The results showed that an T→C mutation in intron 2 at 48500 bp and 3 genotypes (TT, TC and CC) were found in BMP7 gene in Large White Pig, Landrace Pig, Wannan Black Pig and Huoshou Black Pig. The TT was the dominant genotype in Landrace Pig and Large White Pig, its frequency was 73.58%,56% respectively. The CC was the dominant genotype in Wannan Black Pig and Huoshou Pig, and its genotypic frequency was 64.84%ã€67.80% respectively. The polymorphism information content (PIC)was 0.2552ã€0.3108ã€0.3004 and 0.2782 respectively in Landrace, Large White Wannan Black and Huoshou Black population, they were all in average polymorphism situation.(3) the genetic polymorphism parameters of the SNP loci of RBP4 and BMP7 genesχ2 test showed that the gene frequencies and genotype frequencies have reached the Hardy-Weinberg equilibrium (P>0.05) in Landrace,Large White and Wannan Black Pig in RBP4 gene; the gene frequencies and genotype frequencies has reached the Hardy-Weinberg equilibrium (P>0.05) in Large White population, while have not reached in Landrace,Wannan Black Pig and Huoshou Black Pig (P<0.05).(4) the association analysis between A604G locus of RBP4 gene and sows’ litter sizeIn Landrace, Large White and Wannan Black Pig, AA type was the highest inTNB and NBA, followed by AB and BB type. In these three populations, AA type was higher than BB type in TNB and NBA (P<0.05); In Landrace, AA type was higher than AB type in NBA (P<0.05).In Large White and Wannan Black Pig,AB type was higher than BB type in NBA(P<0.05).(5) the association analysis between T48500C locus of BMP7 gene and sows’ litter size:In Landrace, TC type was higher than TT type in NBA (P<0.05). In Wannan Black Pig,CC type was higher than TT type in TNB and NBA (P<0.05). In Huoshou Black Pig, CC type was higher than TT type in TNB (P>0.05).The study show that RBP4 gene was used as candidate genes in MAS which influence litter size in Landrace, Large White and Wannan Black Pig;And BMP7 gene as candidate genes in MAS which influence litter size in Landrace, Wannan Black Pig and Huo Shou pig. |
