The Zinc protein transcription factors Bcl11 a and Bcl11 b play an important regulatory role in the development,differentiation and proliferation of B lymphocyte and T lymphocyte respectively.Previously it has been demonstrated that Bcl11 a and Bcl11 b regulate the expression of a series of downstream genes as nuclear transcription factors,which are responsible for the cell proliferation, cell cycle and immune regulation, with a proven close connection to cancer, and anti-virus function to inhibit the transcription and replication of HIV-1 within the cell.On the account of specific expression of Bcl11 a and Bcl11 b in lymphocytes, and their specific function in immune system are also not clear, we use transgenic mouse and PK15 cell as experimental object, PCV2 as pathogen,mainly by the means of gene deletion,gene overexpression,q PCR,western blot and immunofluorescent assay to study on the function of inhibiting PCV2 multiplication of Bcl11 a and Bcl11 b.At the same time,we use Kunming mice as experimental object, to preliminarily detect the influence of PCV2 on the development of lymphocyte.The main research results and observations are as follows:(1) The mouse immunity decreases significantly after the half deletion of Bcl11a(the Bcl11 a expression level is reduced a half).The PCV2 virus copy number increases significantly, the infection ability of PCV2 on splenic cell is enhanced, PCV2 also cause more serious injury to immune organ such as spleen.(2) The overexpression of Bcl11 a and Bcl11 b in PK15 can significantly inhibit the viral multiplication, and decrease the m RNA, DNA and protein level of PCV2 significantly.(3) Between Bcl11 a, Bcl11 b and PCV2, it does not have obvious colocalization phenomenon.(4)Bcl11a can inhibit the apoptosis of PK15 caused by PCV2 through inhibiting the activity of caspase 3. Bcl11 b can also promote the expression of bcl2, IRF3 and MX1, and cause inhibition of NF-κB expression to inhibit the viral multiplication, then reducing the cell death.The results indicate that the mechanism of Bcl11 a and Bcl11 b inhibit the viral multiplication are different.(5) PCV2 causes a large depletion in the development of lymphocyte, especially have a big impact on B lymphocyte. PCV2 can also inhibit the lymphocyte proliferation in vitro, and have significant inhibition effect on the critical genes involved in the development of lymphocyte. Taken together, PCV2 can inhibit the development and proliferation on the early stage of lymphocyte.
