Rhodiola sp. is a traditional and emerging medicinal plant that grows in hypoxia, cold, intense ultraviolet radiation such harsh environments at elevations of 3,000 m above sea level. It is a valuable research resource as the unique adaptogen activity. This research aimed at the separation and identification of the endophytic fungus from three main Rhodiola sp. which came from two major distribution areas of the Northeast and the Tibetan Plateau of China, their biological diversity and the genetic characteristics were explored respectively. The sterile seedling regeneration system was set up through inducing the callus, subsequently the effects of endophytic fungus on the growth and development status of Rhodiola sp. were studied preliminary.As the investigation showed, it’s extensive to the biodiversity of abundant endophytic fungus were contained in Rhodiola sp.167 kinds of endophytic fungus were separated from 3 types of Rhodiola sp., of which 50 kinds were separated from Rhodiola angusta spreading over 22 genera; 48 kinds were separated from Rhodiola sachalinensis spreading over 28 genera; 69 kinds were separated from Rhodiola crenulata spreading over 27 genera. Fungi strains genetic characteristics analysis showed that they had apparent species specificity and geographic specificity, there were 4 genera owned by the three Rhodiola; Rhodiola angusta、Rhodiola sachalinensis and Rhodiola crenulata particular had 8,16,17 genera respectively; 6 genera existed only in the Rhodiola coming from the Northeast of China and 17 genera existed only in the Rhodiola coming from the Tibetan Plateau of China.The different tissues inducing callus of Rhodiola crenulata was used to set up the plant regeneration system. According to the research, the seed and bud parts were chosen to the sub-experiment materials as the optimal explants. The most effective medium of plant growth substances on callus formation was MS+6-BA 4.0 mg/L+KT 0.1 mg/L+NAA 0.1 mg/L+IAA 0.2 mg/L, the effect degree was 6-BA>KT>2,4-D>NAA>IAA; the most appropriate medium for shoot regeneration was MS+TDZ 0.5 mg/L+NAA 0.5 mg/L, the induction rate was at 63.14%, and the average number of adventitious bud was at 11.4.To screen out the induction activity endophytic fungus which could promote the growth and quality improvement of the Rhodiola, fungi-seedling co-culturing system was established The results showed that Rhodiola angusta, Rhodiola sachalinensis and Rhodiola crenulata respectively had 6,7 and 12 kinds endophytic fungus owing those activities. The fungus which showed growth-promoting obviously belonged to Phialocephala sp., Leptosphaeria sp. and Capronia sp., and fungus that promoted seedling heightening were Coprinellus xanthothrix, Leptosphaeria sp. and Phialophora bubakii, promoted seedling weight gaining was Uncultured Lachnum.
