Atropa belladonna L.is the important medicinal plant species. Recently, with the pharmacological effects of TAs in-depth research and extensive application, there are many researches about it. Elicitor applied to hairy root cultures and their stimulating effects on the accumulation of secondary metabolites is one of the hotspot. In this research, the Atropa belladonna L. hairy root was used as material. Then, the effects of MJ on the growth, adversity physiological characteristics and TAs accumulation of in Atropa belladonna L. hairy roots were studied. The main results were as follows:1. The effects of medium types on hairy root growth and TAs production were studied in suspension culture of Atropa belladonna L. hairy root. The biomass of Atropa belladonna L. hairy root in B5 medium and MS medium was significantly higher than that in N6 medium, and there was no significant difference between the two mediums. In B5 medium, the production of scopolamine and hyoscyamine were significantly higher than that in MS medium. The results showed that B5 medium was best not only for hairy root growth but also for the accumulation of alkaloids.2. The effects of MJ on the growth and adversity physiological characteristics of in Atropa belladonna L. hairy roots were studied. The results demonstrated that MJ had a negative effect on the growth of hairy roots. Different concentrations of MJ were induced, hairy roots appear yellowing and senescence. After treated by MJ, the contents of O2-. soluble protein, MDA, pro of Atropa belladonna L. hairy roots increased in different degrees or increase firstly and then decrease. The POD activity in 10μmol·L-1MJ group was highest. With the increase of MJ concentration, POD activity decreased. The SOD activity in 20μmol·L-1 MJ group was highest, which the groups with MJ treatment were significantly higher than control. The CAT activity in 40μmol·L-11MJ group was highest, which was significantly higher than other groups. The results illustrate, MJ affects Atropa belladonnah. hairy roots oxidase activity.4.The results show that, in Atropa belladonna L. hairy roots, MJ could significantly promote the accumulation of TAs. MJ feeding(10μmol·L-1,12d) resulted in the highest content of hyoscyamine, and the content of scopolamine in 30μmol·L-1 MJ group was the highest. Due to the influence of hairy root biomass, the hyoscyamine yield in 10, 20μmol·L01 MJ group has no significant difference with the control. The MJ-treated samples displayed increase in h6h and pmtgene expression.
