As the pharmacological activity research of Polygonatum sibiricum is unceasingly thorough, the demand is increasing. But the wild resources cannot satisfy the demand of people, so Polygonatum sibiricum breeding mainly adopts rhizome reproduction. However,prolonged vegetative propagation cause degradation of Polygonatum sibiricum species, pests increase, the eyes of scholars gradually shifted to seed propagation. The Polygonatum sibiricum seed with comprehensive dormancy, current Polygonatum sibiricum seed hypocotyl dormancy breaking technology is now mature. But the basic is the single factor analyses, this paper through the comprehensive analysis of four factors(GA3 concentration, soaking temperature, soaking time, disinfectants) three levels orthogonal experiment, draw the optimal solution. Now there is a few studies about epicotyls’ dormant of Polygonatum sibiricum seed, and the related technologies are not mature enough.. So in this paper, by studying the different concentrations of 6-BA and different cultivation substrate influence on Polygonatum sibiricum primary root emergence, to obtain the concentration and growing media, which can provide an experimental basis of Polygonatum sibiricum seedling formation.The main findings are as follows:1. Which combined GA3 comprehensive, seed soaking temperature, soaking time,disinfectants four factors, obtained that the best pretreatment conditions of Polygonatum sibiricum seed germination is using 100 mg / L of GA3 normal temperature(average temperature is about 20 ° C),soaking with 12 h, disinfecting with H2O2.Among them,the soaking temperature is the major factor of Polygonatum sibiricum seeds germination, soaking time is next, the GA3 concentration and disinfectant effect had the smallest function on the results.2. The study obtained that with the rise of 6-BA concentration that the emergence rate and emergence index of Polygonatum sibiricum primary roots have a tend which first increased and then decreased. After the treatment with 100 mg / L of 6-BA, it could very significantly(p<0.01) increase emergence rate and emergence index of Polygonatum sibiricum primary roots, after the treatment with 150 mg / L of 6-BA, it could significantly(p<0.05) increase emergence rate and emergence index of Polygonatum sibiricum primary roots,and 100 mg / L, 150 mg / L of 6-BA are the optimal concentration to deal with Polygonatumsibiricum primary roots. 250 mg / L of 6-BA could inhibit the emergence of Polygonatum sibiricum primary roots.3. The study obtained that nutrient soil: vermiculite(2: 1) could significantly(p<0.05)increase the emergence rate and emergence index of Polygonatum sibiricum primary roots,the emergence rate and emergence index reached 52.32%, 4.95.
