The Escherichia coli disease of rabbits can lead to the diarrhea, especially the rabbits from 4-week-old to 8-week-old, it is an serious disease of feeding rabbits. Because of the high incidence, high mortality, big loss and the low level of cure, people who are in the area of feeding rabbits from all of the world care about the E. coli disease of Rabbits.The main pathogenic factors of E. coli are serotype, endotoxin and adsorption.E. coli own so many serotypes leading to diseases of people or animals, and the serotypes vary from one plant to another in one epidemic area. The lipopo-lysaccharide from the outer membrane of E. coli is the endotoxin of E. coli, and the endotoxin is the main factor of the pathogenic of E. coli. The pili of E. coli have something with adsorption which is treated as the premise condition.After cracking, E.coli in rabbit body can release the lipopo-lysaccharide recognized and binded to by TLR4 from TLRs, then they cause a series of signal transduction especially the TLR4/NF-κB pathways and produce all kinds of inflammatory mediators or factors, for example, IL-1B, IL-6, IL-8, TNF-A. After that, the inflammatory mediators or inflammatory factors participate in the inflammatory process of E. coli infections.This experiment can implementate positioning of quantitative analysis of the expressions of substantive organs’ TLR4 of rabbits with E. coli in different periods via immunohistochemical method, so that we can get the TLR4’s distribution rules in different organs or times and the sensitivities of different substantive organs to E. coli. The TLR4 expression quantity can be illustrated from the side the number of inflammation factors produced by each essential organ in the process of anti-infection, the importance of the each essential organ and the degree of its infection. The research can provide certain theoretical basis for the E. coli disease of young rabbits.The positive points of heart distribute in myocardial fibers; TLR4 expresses mainly in liver cells, coop cells, endothelial cells, but we can find the expressions of TLR4 in bile duct epithelial cells and other cells of the cytoplasm and cell membrane; Spleen’s positive points are mainly distributed in the white pulp lymph nodules and lymphoid tissue sheath surrounding arteries, and also we can find the expressions of the receptors around the trabeculars; Lung’s positive points are mainly distributed in the alveolar ducts, alveolar around, but also distributed in a small amount of fine bronchial mucosa; The kidney’s positive points are mainly distributed in the renal tubules surrounding capillaries, renal tubular epithelial, are also expressed in the glomerulus.Both healthy animals and animals in disease, heart, liver, spleen, lung and kidney can be found the expressions of TLR4, but the TLR4 expression quantities exist differences. The number of the expressions of TLR4 in spleen, kidney volume significantly higher than the amount of TLR4 expressions in the heart, lung and liver. And their differences of different animals at the same stage of the onset lead to the slightly different quantities of expressions of TLR4. At the different onset periods, the essences of the different organs in the process of anti-infection play important roles. In the first day, kidney, liver and lung’s expressions of TLR4 reach the highest and the very three organs play serious roles in the body to resist invasion of E. coli. In the third day, spleen’s expressions of TLR4 reach the highest and spleen plays a serious role. In the the fifth day of onset, the number of the expressions of TLR4 in the heart reaches the highest and heart plays a serious role.As for the TLR4’s expression in the heart, the fifth day of E. coli infection group and normal control group, 1, 3, 7 days E. coli infection group exist significant differences(P <0.05); As for the TLR4’s expression in the liver, the first day of E. coli infection group and normal control group, 3, 5, 7 days E. coli infection group exist significant differences(P <0.05); As for the TLR4’s expression in the spleen, the second day of E. coli infection group and normal control group, 1, 5, 7 days E. coli infection group exist significant differences(P <0.05); As for the TLR4’s expression in the lung, the first day of E. coli infection group and normal control group, 3,5, 7 days E. coli infection group exist significant differences(P <0.05); As for the TLR4’s expression in the kidney, the first and fifth day of E. coli infection group and normal control group, 3, 7 days E. coli infection group exist significant differences(P <0.05).Because of the different numbers of TLR4 receptors in heart, liver, spleen, lung, kidney, as for the expressions of TLR4 receptors in the same onset period, there are differences in the substantive organs.By means of the peak quantities of the TLR4 expressions of organs and peak periods, we can calculate the sensitivity of each substantive organ in the process of the invasion of the E. coli: kidney >liver > lung> spleen > heart.
