Laminitis is a common motor system disease affecting both horses and ponies. In the current study, we use experimental model of acute laminitis induced by OF to detect the changing of LPS, ADPN, insulin, glucose, IL-6, p38 MAPK, 5-HT, ET-1, TXB2 and NO in plasma and the pathological change in hoof, investigate the pathogenesis of acute laminitis, search for biomarkers that can be used in early detection and rapid diagnosis, and provided the theoretical basises for the treatment of laminitis in future.21 Mongolian horses were divided into experimental and control groups. The bolus induction dose of 10 g/kg bwt OF that was dissolved in water and administered by nasogastric tube to experimental horses; Control horses were administered equal magnitude distilled water. Euthanasia were performed to horses which reached the 3rd grade that evaluated by Obel Grade, then took out laminar for HE-stained and PAS-stained.The results showed that 11 horses of experimental group appeared the clinical symptomses such as fervescence, respiration and heart rate acceleration, lameness, and diarrhea, and reached 1st grade of lameness after 8-20 h of perfusion and reached 2nd grade of lameness after 24-56 h of perfusion.Along with the 2nd grade of lameness,the rate of respiration and heart of horses increased, the p H of faeces presented a minimum value at 20 h, and the rectal temperature reached a peak at 40 h. All of the horses of experimental group occurred diarrhea during 12-24 h and remitted after 60 h; however, the horses of the control didn’t present discomfort.We found that the 3rd phalanx of the horses in OF treatment spinned through X-ray and some histological changes such as inflammatory cell infiltration, basic dyestuff gathered around blood vessels, but the similar results were not found in the horses of the control.The plasma concentration of LPS was significantly increased from 8 h and reach a peak at 16 h before decreasing back to control levels by 40 h, which was significantly increased compared with 0 h(P < 0.01); the plasma concentration of ADPN and insulin were significantly increased at 4 h, 8 h and reached the nadir and peak at 36 h respectively, which were all significantly increased compared with 0 h(P < 0.01); the plasma glucose level was significantly increased at 4 h till 16 h and reached a peak at 4 h, which was significantly increased compared with 0 h(P < 0.01); the plasma concentration of IL-6 was significantly increased at 24 h till 68 h and reached a peak at 32 h, which was significantly increase compared with 0 h(P < 0.01); the plasma concentration of p38 MAPK was significantly increased from 8 h and reached a peak at 16 h and which was greater compared with 0 h(P < 0.01); the plasma concentration of 5-HT, ET-1, and TXB2 were greater at from 4 h, 8 h, and 8 h till 60 h and reached the peak at 24 h, 24 h, and 28 h respectively, which were all significantly increased compared with 0 h(P < 0.01); the plasma concentration of NO was significantly increased from 8 h till 60 h and reached a peak at 32 h, which had a significant increase compared with 0 h(P < 0.01).The results of the present study show that increasing absorption of LPS cause some material secretion disorder such as IL-6, ET-1, ADPN and insulin, the trend of the changes of LPS, ADPN, insulin, IL-6, 5-HT, ET-1, TXB2 and NO matched with the trend of the changes of the course of laminitis.
