Identification Of Hybrid Rapeseed And Construction Of Varieties ID By SSR Markers

Rape belongs to cruciferous brassica plant, which is an important oil cash crop. Rape is widely distributed in china, with an average annual acreage and total production accounting for about 1/3 of the world. Hybrid rapeseed is warmly received by seed companies and farmers due to its high yield, stress resistance and wide adaptability. With the rapid development of hybrid rapeseed and breeding technology, a lot of new varieties had emerged, so are the counterfeits. It results in disadvantageous effects for promotion of rape seed quality management and protection of variety rights. Therefore, it is necessary to establish an accurate, reliable technical method to identify rape varieties. In other hands, the construction scheme for rape variety ID was designed, and bar code or QR code of the variety ID was structured for 49 rape varieties. The main results were as follows:1. To properly differentiate hybrid rapeseed varieties,33 pairs of SSR primers evenly distributed on 19 chromosomes were screened. Meanwhile, the sampling, testing and interpretation were optimized. Our results revealed that there were three patterns of SSR genotype between samples:the identical type, the suspected identical type and the distinct type. After investigate the SSR pattern of 12 hybrid rapeseed varieties, we found that there were 4-22 distinct events among them. The half-blood varieties with same father exhibited 4-16 distinct events in all of 33 testing, and the half-blood varieties with same mother exhibited 7-18 distinct events. Further, the marketing varieties were examined, and 12-20 distinct events were found between two varieties.After testy all 12 varieties, we found 0～2 distinct events may occurred in single variety. Therefore, the sample could be recognized as the same variety if the distinct events were less than 2, while, they are recognized as the different varieties.2. Making use of the method to detect 5 blind samples from the seed manage department, our method had been proven by the culturing identification and blind identification.3. Referring to the construction of human ID, the commodity information, molecular fingerprinting information and the specific genetic information were combined to construct rape variety ID.49 rape varieties ID were constructed according to the scheme, and were converted into the corresponding bar code and QR code by specific software.
