Labiatae catnip nepeta is a pernennial herb.also known as hong fok,nepeta fake,fakesoviet union and so on,all the grass aroma,there is flooding control and insecticidefunction.Catnip can be medicinal,lesves and flowers open end can treat cold sheadaches and scalp itching.Is a great value for aromatic plants.In this paper,on the one hand the use of plant tissue culture techniques toestablish catnip regeneration system,on the other hand the volatile oil extracted by steam distillation,using GC-MS technique catnip to analyze the chemical composition of volatile oil,volatile oil and catnip vitro antibacterial,toxicity evaluation and the role of mosquitoes driving off to studyaimed cultivation,development and utilization of catnip provide a theoretical basis and technical support.Reserch results are as follows:1.Catnip established regeneration system.This article regeneration system were established to catnip leaves,shoot tips,bud,stem catnip as explants,the results showed that:in the scope of the study to bud as explants for callus induction contamination lowest rate 13.3%,mortality rate of 10.9%,the optinal callus explant to bud;optimal medium for inducing adventitious buds catnip for 6-BA 2.5mg·L-1+NAA1.0 mg·L-1,catnip-induced the best medium for adventitious should be 1.0 mg-L-1+NAA1.0 mg·L-1+6-BA 0.6 mg-L-1;carbon concentration of 3%sucrose best.2.By steam distillation,in order to influence the volatile oil extracted from catnip four mainfactors:the sample size of the soaking time,add water,extraction time or thogonal experiment,the optimum screening of volatile oil extracted from catnip.The results show that:in the study area,soak time of 10 hours,plus 10 times the amout of water quality catnip,extraction time of 12hours,herbs particle diameter is preferably a medium,its volatile oils have the highest rate of 3.25mL · kg-1.3. Analysis of catnip chemical composition of volatile oil mass spectrometry(GC-MS)-using gas chromatography.The results show that:the volatile oil indentified 40 kinds of compunds,which contains sesquiterpene substance 24 species,accounting for 58.44% of the total essential oils,mainly (E)-β-farnesene (7.40%), trans-β-law farnesene (5.36%), a-terpineol (4.25%), a-pinene (3.63%), eucalyptol enol (3.13%), cineol (2.57%) and the like. Indoles a substance,2-methylindoline.Aromatic compound,3-hydroxy-4-methoxy toluene.14 kinds of aliphatic compounds,representing 41.11%,mainly E,Z-nepetalactone false(17.14%),Z,E-false nepetalactone(12.02%),pepper ester(0.65%%)and the like,False nepatalactone(29.16%)is one of the most volatile catnip oil components.4. Cup and saucer was studied by volatile oil of catnip common fungal and bacterial inhibition and determination of minimum inhibitory concentration. The results showed that:volatile oil of catnip fungi and bacteria have a certain extent. Inhibition of bacteria showed that:Catnip volatile oil are stronger inhibitory effect on bacteria and volatile oil of catnip inhibition of E. coli and B. subtilis bacteria quadruple superior to inhibition; volatile oil of catnip quadruple coli bacteria the minimum inhibitory concentration (MIC) value of 50% for E. coli and B. subtilis MIC values of 25%. Inhibition of fungal indicate: volatile oil of catnip Rhizopus inhibition was significantly better than the inhibition of Aspergillus niger, Rhizopus which MIC values of 6.25%; while Aspergillus niger MIC is 75%.5.By acute oral toxicity test and skin toxicity evaluation of toxicological tests to make catnip. Acute oral toxicity experiments showed that:The number of dead mice in creased with increasing dose of volatile oil,after the mice were fed with different degrees of poisoning a week living memory mice were normal activities.Experimental results show that the volatile oil of catnip LD50 of 41.385g· kg-1 of median lethal dose in mice is much greater than 15g·kg-1,in accordance with national standards for acute toxicity rating criteria,are non-txic substances.Toxicology results showed that skin:Skin irritation test,no intact skin and damaged skin redness,blistering and other irritating greactions.In the sensitization test,after giving allergic guineapigs showed no symptoms,according to skin irritation,allergic reactions score sheet determination of volatile oil of catnip non-irritating,non-allergenic,security is good.6.Study the volatile oil of catnip aedes albopictus and housefly repellent,contact action.Apply repellent skin test by measuring aedes albopictus repellent effect,the results showed that:Catnip volatile oil has good msquito repellent,insectrepellent effect was enhanced with increasing dose,mosquito mice stopped volatile oil painting drop significantly smaller than the number of blank control group. When the dose of volatile oil painting 1.12μL · cm-2,its repellent effect can be achieved by six god toilet water repellent similar effect,are effective protection for about 8h,at this dose is less than the national standard of 1.5μL· cm-2,the effective protection time can be up to 10.6h,higher than the existing similar products,insect repellent effect Study membrane catnip volatile oil plus flies on contact action,results showed that water extraction of volatile oil of catnip housefly dose dependent increase in the calculated half of the volatile oil of catnip housefly lethal dose of 3.63%.showing the effect of low levels of highly efficient tag.
