Correlation Between Extracellular Enzyme Activity Of Metarhizium Anisopliae And Virulence And The Study Of Application Technology

The extracellular protease activity,chitinase activity, lipase activity of 6 Metarhizium anisopliaeand M11-8-7 、 M7-5-7 、 M7-10 、 M6-448 、 M8-5-83 and M8-6-291, its correlation with the toxicity of Tenebrio molitor L. the Folin-phenol Reagent method,DNS method and Acid-Base Titration method were analyzed. The results showed that there was an obvious difference in production of extracellar protease among different strains and there was an obvious liner correlation between extracellular protease activity and virulence.The equation is y = 47.79-12.19 x,R2 = 0.8978.But chitinase and lipase do not have the obvious liner crrelation with virulence. With the results of the virulence assay,we filter out two strains of Tenebrio molitor L. with high virulence and high extracellular enzyme activity Metarhizium anisopliae, they are M11-8-7 and M7-10.We filter out one strain with high virulence,good effect of fermentation and difficult to pollute Metarhizium anisopliae,it is M11-8-7. Then we did the field simulation experiments. 450 billion spores/mu’s Metarhizium anisopliae had good lethal effect to Carposina sasakii.5, 10 trillion spores/mu’s increase in the fresh weight yield of peanuts were 32.98% and 36.8%.The effect of pest control were 56.25% and 87.5% and the control effect were 52.08% and 76.77%. Applying Metarhizium anisopliae is influenced by Animal manure. The chicken manure and the sheep manure made the Metarhizium anisopliae’s control effect is not obvious.The Metarhizium anisopliae’s control effect with the cow dung and the water were 66.08% and 74.69%.
